基于TCGA数据集分析mRNA在子宫内膜癌中差异表达及对预后影响 被引量:3

摘要 目的基于TCGA数据集分析mRNA在子宫内膜癌中的差异表达,建立mRNA差异表达与子宫内膜癌预后的关联。方法提取TCGA数据库中子宫内膜癌样本的mRNA表达量及临床数据资料,采用edgeR算法比较正常组织与子宫内膜癌组织的mRNA表达量,通过mRNA差异表达和临床存活时间作生存分析,寻找对子宫内膜癌预后产生影响的mRNA。结果正常组织与子宫内膜癌组织之间共发现4 429个差异表达mRNA,从中选取前1 000个差异表达mRNA进行GO富集分析、KEGG富集分析及蛋白互作网络分析,筛选出20例hub基因。通过生存分析发现,7个差异表达mRNA(ASPM、AURKA、BUB1、CDCA8、KIF2C、TOP2A、UBE2C)与子宫内膜癌患者存活时间相关(P <0. 05)。结论正常组织和子宫内膜癌组织之间的ASPM、AURKA、BUB1、CDCA8、KIF2C、TOP2A、UBE2C 7个差异表达mRNA与子宫内膜癌患者预后相关,对mRNA在子宫内膜癌领域的研究具有参考价值。 Objective Differential mRNA expression in endometrial cancer was analyzed based on TCGA data set to establish the correlation between differential mRNA expression and prognosis of endometrial cancer. Methods The mRNA expression levels and clinical data of endometrial cancer samples from TCGA database were extracted,and mRNA expression levels of normal tissues and endometrial cancer tissues were compared by edgeR algorithm. Survival analysis was conducted through differential mRNA expression and clinical survival time to find the mRNA that had an impact on the prognosis of endometrial cancer. Results A total of 4 429 differentially expressed mrnas were found between normal tissues and endometrial cancer tissues,among which the first 1 000 differentially expressed mrnas were selected for GO enrichment analysis,KEGG enrichment analysis and protein interaction network analysis to screen20 cases of hub genes. Survival analysis showed that seven differentially expressed mrnas( ASPM,AURKA,BUB1,CDCA8,KIF2 C,TOP2 A,and UBE2 C) were associated with survival in patients with endometrial cancer( P < 0. 05).
