核桃叶片多酚类物质积累及相关酶活性分析 被引量:1

摘要 以不同品种核桃中林5号、香玲、礼品1号、礼品2号叶片为试材,研究不同品种核桃叶片生长发育中多酚类物质积累及相关酶活性的变化。结果表明,核桃叶片多酚类物质的含量在其生长周期呈现一定的变化规律,5月25日至7月5日下降,7月5日至8月18日上升,8月18日至9月4日有所下降;HPLC鉴定出没食子酸、儿茶素、绿原酸、咖啡酸、丁香酸、丁香醛、对香豆酸、阿魏酸、芦丁、杨梅素、胡桃醌、槲皮素共12种单体酚,含量最高的为芦丁,其次为儿茶素,含量较低的有没食子酸、丁香醛等,核桃叶片中大多数单体酚在7、9月含量较低,在5、8月含量较高;酶标仪测定的3-脱氧-阿拉伯庚酮酸糖-7-磷酸合酶(DAHPS)、苯丙氨酸解氨酶(PAL)、肉桂酸-4-羟化酶(C4H)、分支酸变位酶(CM)、柠檬酸合酶(CS)、查尔酮合成酶(CHS)共6种酶活中,大部分酶的活性随着时间的推移而升高,活性最高的为3-脱氧-阿拉伯庚酮酸糖-7-磷酸合酶(DAHPS),其次是肉桂酸-4-羟化酶(C4H),最低的为柠檬酸合酶(CS)。 The leaves of different walnut varieties(Zhonglin 5,Xiangling,Lipin 1,Lipin 2)were used as experimental materials to study the accumulation of polyphenols and related enzyme activities in the growth and development of walnut leaves.The results showed that the content of polyphenols in walnut leaves changed regularly during its growth cycle,which decreased from May 25 to July 5,increased from July 5 to August 18,and decreased from August 18 to September 4.HPLC identified 12 kinds of monomeric phenols,including gallic acid,catechin,chlorogenic acid,caffeic acid,eugenic acid,eugenal,p-coumaric acid,ferulic acid,rutin,myricetin,walnut quinone and quercetin.The highest content was rutin,followed by catechins.The lower content was gallic acid,clove aldehyde,and so on.The content of most monomer phenols in walnut leaves was lower in July and September,but was higher in May and August.There were 6 kinds of enzyme activities determined by the microplate reader,including 3-deoxy-arabinophenone glucose-7-phosphatase synthase(DAHPS),phenylalanine ammonia-lyase(PAL),cinnamate-4-hydroxylase(C4H),branched acid mutase(CM),citric acid synthase(CS)and chalone synthase(CHS).Most of the enzyme activities increased with delaying of the time,and the highest activity was DAHPS,followed by C4H,the lowest was CS.
