摘要
研究新疆哈密大枣中环磷酸腺苷(cyclic adenosine monophosphate,cAMP)的分离纯化工艺。以吸附解吸率为衡量指标,通过对比4种大孔树脂的静态吸附与解吸,确定纯化哈密大枣提取液中cAMP的最佳树脂;通过大孔树脂动态吸附与洗脱,考察上样浓度、p H值、流速等因素,确定cAMP分离纯化的最佳工艺。结果表明:HZ-802型树脂纯化效果较好,动态吸附的最佳条件为:上样体积220 mL,上样液流速2.0 m L/min,上样液浓度20μg/mL,上样液pH 5;动态解析的最佳条件:洗脱液40%乙醇,洗脱液体积120 mL,洗脱液流速为3 mL/min;经纯化后得到环磷酸腺苷浓度为38.24μg/mL,冷冻干燥后粗提物中cAMP含量为0.10%。
The separation and purification technology of cAMP from Xinjiang Hami jujube was studied. By comparing the static adsorption and desorption of four macroporous resins,the best resin for purification of cAMP in the extract of Hami jujube was determined. By dynamic adsorption and elution of macroporous resin,the factors such as sample concentration,pH and flow rate were investigated to determine the best purification process of cAMP. The results showed that HZ-802 resin had better purification effect. The optimum conditions for dynamic adsorption were as follows:sample volume 220 mL,flow rate 2.0 mL/min,concentration 20 μg/mL and pH 5 of sample solution. The optimum conditions for dynamic analysis were as follows:40 % ethanol in eluent,120 mL in eluent and 3 mL/min in eluent. After purification,the concentration of cAMP was 38.24 μg/mL,and the content of c AMP in the crude extract was 0.10 % after freeze-drying.
