摘要
目的探讨结直肠癌(CRC)组织中微小RNA-137(miR-137)、Notch1及Delta样配体4(DLL4)表达变化及临床意义。方法选取CRC患者100例,术中收集新鲜CRC组织及距肿瘤边缘>3 cm的正常组织。采用荧光定量PCR法检测CRC组织及癌旁正常组织中miR-137表达,用免疫组化SP法检测CRC组织及癌旁正常组织中Notch、DLL4蛋白表达。分析结直肠癌组织中miR-137、Notch1、DLL4表达与患者临床病理特征的关系,Pearson线性相关分析CRC组织中miR-137与Notch1、DLL4蛋白表达的相关性。结果CRC组织与癌旁正常组织中miR-137相对表达量分别为0.79±0.24、3.61±0.69,CRC组织中miR-137相对表达量低于癌旁正常组织(t=38.601,P=0.000)。CRC组织与癌旁正常组织中Notch1蛋白阳性表达分别为47例(47%)、5例(5%),CRC组织中Notch1蛋白阳性表达率高于癌旁正常组织(χ2=45.842,P=0.000)。CRC组织与癌旁正常组织中DLL4蛋白阳性表达分别为81例(81%)、29例(29%),CRC组织中DLL4蛋白阳性表达率高于癌旁正常组织(χ2=50.731,P=0.000)。CRC组织中miR-137表达与肿瘤分期有关(P<0.05),与CRC患者其他临床病理特征无关(P均>0.05);Notch1、DLL4蛋白阳性表达与肿瘤分期、淋巴结转移及远处转移有关(P均<0.05),与性别、年龄、肿瘤大小、肿瘤位置、组织分化程度无关(P均>0.05)。CRC组织中miR-137的相对表达量与Notch1、DLL4蛋白相对表达量呈负相关(r分别为-0.654、-0.612,P均<0.05)。结论CRC组织中miR-137呈低表达,而Notch1、DLL4呈高表达,三者可能协同参与CRC的发生发展。
Objective To investigate the expression and clinical significance of microRNA(miR)-137,Notch1,and Delta-like ligand 4(DLL4)in the colorectal cancer(CRC)tissues.Methods The fresh CRC tissues and normal tissues>3cm away from cancer tissues of 100 patients with CRC were collected.Fluorescence quantitative PCR was used to detect the expression of miR-137 in the CRC tissues and adjacent normal tissues.The expression of Notch1 and DLL4 protein was detected by immunohistochemical SP.The relationship between the expression of miR-137,Notch1,and DLL4 and the clinicopathological features in the colorectal cancer tissues was analyzed.Results The relative expression of miR-137 was 0.79±0.24 and 3.61±0.69 in the CRC tissues and adjacent normal tissues,respectively.The relative expression of miR-137 in the CRC tissues was lower than that in adjacent normal tissues(t=38.601,P=0.000).The positive expression rate of Notch1 protein in the CRC tissues and adjacent normal tissues were 47%and 5%,respectively.The positive expression rate of Notch1 protein in the CRC tissues was higher than that in the adjacent normal tissues(χ2=45.842,P=0.000).The positive expression of DLL4 protein in CRC tissues and adjacent normal tissues were 81%and 29%,respectively.The positive expression rate of DLL4 protein in the CRC tissues was higher than that in the adjacent normal tissues(χ2=50.731,P=0.000).The expression of miR-137 in the cancer tissues was related to tumor stage(P<0.05),but not to the other clinicopathological features of CRC patients(all P>0.05).The positive expression of Notch1 and DLL4 was related to tumor stage,lymph node metastasis,and distant metastasis(all P<0.05),but not to sex,age,tumor size,tumor location or tumor differentiation(all P>0.05).The relative expression of microR-137 was negatively correlated with Notch1 and DLL4 in the CRC tissues(r=-0.654,-0.612,both P<0.05).Conclusion The expression of miR-137 in the CRC tissues decreases,while Notch1 and DLL4 increase,and these three factors may be involved in the occurrence
