摘要
目的研究A型肉毒毒素(botulinum toxin A,BTXA)对增生性瘢痕组织成纤维细胞的作用及其bFGF、caspase-3及Bcl-2表达的影响。方法实验用的成纤维细胞来源于本科室患者的增生性瘢痕的组织块培养获得。在体外用含有0.2~1.6 U/ml浓度的BTXA的培养液孵育增生性瘢痕的成纤维细胞,采用MTT法检测细胞的存活率,吖啶橙-溴化乙锭(AO/EtBr)双染色的方法检测细胞形态;同时将浓度分别为0.2、0.4、0.8、1.6 U/ml A型肉毒毒素的培养液孵育24 h的增生性瘢痕成纤细胞用RT-PCR及Western blotting检测其bFGF、caspase-3及Bcl-2 mRNA及蛋白的表达情况。结果体外培养的瘢痕成纤维细胞的生长受到A型肉毒毒素的抑制而导致凋亡,并且这种抑制作用随着药物浓度的增高而加强。随着A型肉毒毒素浓度的增加bFGF和Bcl-2mRNA及蛋白的表达明显下降,caspase-3水平明显升高(P<0.05)。结论BTXA抑制瘢痕增生的机制可能与下调bFGF和Bcl-2的表达,上调caspase-3表达有关。
Objective To investigate the effects of botulinum toxin A(BTXA)on the expressions of bFGF,caspase-3 and Bcl-2 in fibroblasts of hypertrophic scar tissue.Methods The fibroblasts from hypertrophic scar tissues were cultured by tissue block method.BTXA of different concentrations was used to human fibroblasts culture in vitro.The cell survival rate was detected by MTT,and cell morphology was detected by acridine orange-ethidium bromide(AO/EtBr)double staining.Moreover the fibroblasts from hypertrophic scar tissues were cultured in the medium with concentrations of 0.2,0.4,0.8,1.6U/ml BTXA for 24 hours,then the expressions of BTXA on bFGF,caspase-3 and bcl-2 mRNA and protein were detected by RT-PCR and Western Blot.Results BTXA could significantly inhibit the proliferation of hypertrophic scar fibroblasts in a dose-dependent manner.The expression levels of bFGF,bcl-2 mRNA and protein were significantly decreased,however,the expression levels of caspase-3 were significantly increased(P<0.05).Conclusion The action mechanism of BTXA in inhibiting scar hyperplasia may be related to down-regulating the expressions of bFGF and bcl-2 and up-regulating the expression of caspase-3.
