隐球菌性脑膜炎小鼠VEGFR与CD93和CD200的表达及临床意义

Expression and clinical significance of VEGFR, CD93 and CD200 in mice with cryptococcal meningitis

摘要 目的探究血管内皮细胞生长因子受体(Vascular Endothelial Growth Factor Receptor,VEGFR)、外泌体CD93、外泌体CD200在小鼠隐球菌性脑膜炎中的表达及临床意义。方法选择健康成年小鼠60只,随机分为研究组40只和对照组20只。研究组小鼠进行免疫抑制,后尾静脉缓慢推注新鲜新生隐球菌,对照组注射等剂量生理盐水。检测并比较两组小鼠脑组织VEGFR和血清炎性因子、CD93、CD200水平,并分析人肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)、IL-17与外泌体CD93、外泌体CD200间的相关性。结果建模后4 h研究组小鼠脑组织VEGFR1和VEGFR2水平升高(P<0.001),而第12 h和24 h均低于对照组和研究组4 h(P<0.001),研究组小鼠血清CD93、CD200蛋白分别为(85.21±21.53)pg/ml、(221.83±84.25)pg/ml高于对照组(P<0.05)。两组的IL-4和TNF-α差异无统计学意义,研究组IL-17为(17.32±6.32)pg/ml高于对照组(P<0.05),研究组IFN-γ为(34.64±9.46)pg/ml低于对照组(P<0.05)。CD93、CD200与IFN-γ呈负相关性(P<0.05),与IL-17呈正相关性(P<0.05)。结论低水平的VEGFR可能有助于缓解隐球菌性脑膜炎脑水肿,而CD93、CD200可通过调节Th1和Th2免疫途径清除病原体。 OBJECTIVE To explore the expression and clinical significance of vascular endothelial growth factor receptor(VEGFR), CD93 and CD200 in mice with cryptococcal meningitis. METHODS Forty healthy adult mice were randomly divided into the study group and the control group. The mice in the study group were immunosuppressed, and injected with Cryptococcus neoformans slowly into the posterior caudal vein, whereas the mice in the control group were injected with an equivalent amount of saline. The levels of VEGFR, serum inflammatory factors, CD93 and CD200 were detected and compared between the two groups. The correlation between levels of TNF-α, IFN-γ, IL-4 and IL-17 and levels of CD93 and CD200 was analyzed. RESULTS The levels of VEGFR1 and VEGFR2 in the brain tissue of mice in the study group increased 4 hours after modeling(P<0.001), which was lower than that in the control group at the same time point, as well as significantly lower than that in the control group 12 and 24 hours after modeling(P<0.001). The relative protein content of CD93(85.21±21.53)pg/ml and CD200(221.83±84.25)pg/ml in the study group was significantly higher than that in the control group(P<0.05). There was no significant difference in the levels of IL-4 and TNF-α between the two groups. The relative expression of IL-17 in the study group was(17.32±6.32)pg/ml, which was significantly higher than that in the control group(P<0.05);IFN-γ in the study group was(34.64±9.46)pg/ml, and was significantly lower than that in the control group(P<0.05). Levels of CD93 and CD200 were negatively correlated with IFN-γ(P<0.05), and positively correlated with IL-7(P<0.05). CONCLUSION Low levels of vascular endothelial growth factor receptor may help to alleviate cryptococcal meningitis encephaledema. CD93 and CD200 can eliminate pathogens by regulating Th1 and Th2 immune pathways.
