急性心肌梗死患者循环内皮细胞相关基因的生物信息学分析

摘要 目的应用生物信息学法对急性心肌梗死(AMI)患者外周血循环内皮细胞的相关基因进行分析。方法从基因表达综合数据库(GEO)中下载基因芯片数据集GSE66360利用GE02R、DAVID及STRING等分析平台筛选差异表达基因,并进行基因本体(GO)分析、信号通路(KEGG)分析及蛋白-蛋白相互作用网络(PPI)分析,应用Cytoscape软件进行模块分析并筛选核心基因。结果共筛选出2个下调基因和80个上调基因差异表达基因主要富集于免疫应答、炎症反应、碳水化合物类结合、细胞外基质、细胞因子-细胞因子受体相互作用等生物学过程;识别出的9个核心基因中,IL1B、SLC11A1及LILRB2为显著上调的差异基因,CCR2为下调基因。结论AMI时循环内皮细胞出现表达变化的基因功能主要富集在免疫应答、炎症反应、碳水化合物类结合、细胞外基质、细胞因子-细胞因子受体相互作用等生物学过程,其中显著变化的核心基因IL1B、SLC11A1、LILRB2及CCR2可能发挥着重要作用,可能是AMI的预防和治疗的靶点。 Objective To analyzed the related genes of endothelial cells in peripheral blood circulation of patients with acute myocardial infarction(AMI)by bioinformatics.Methods The gene chip data set(GSE66360)was downloaded from the gene expression comprehensive database(GEO),and differential expression genes were screened by analysis platforms of GEO2R,DAVID and STRING.Gene Ontology(GO)analysis,signal pathway(KEGG)analysis and protein-protein interaction network(PPI)analysis were carried out.The module analysis and core genes were screened by Cycloscape.Results Two down-regulated genes and 80 up-regulated genes were screened out,and the differentially expressed genes were mainly enriched in biological processes such as immune response,inflammatory response,carbohydrate binding,extracellular matrix and cytokine-cytokine receptor interaction.Among the nine identified core genes,IL1B,SLC11A1 and LILRB2 were significantly up-regulated differentially genes,and CCR2 was down regulated gene.Conclusions In AMI patients,the gene functions of expression changes of circulating endothelial cells were mainly enriched in biological processes such as immune response,inflammatory response,carbohydrate binding,extracellular matrix and cytokine-cytokine receptor interaction.The core genes with significant changes,such as IL1B,SLC11A1,LILRB2 and CCR2,may play important roles and may be the targets of prevention and treatment of AMI.
