摘要
细胞外基质(ECM)包含复杂的蛋白质环境和多种生长因子,为细胞提供机械、生物物理和生物化学信号,具有组织器官特异性。器官衍生的脱细胞ECM(dECM)是目前最具仿生生物学的材料,这一特点使其在3D生物打印应用中成为热门材料。笔者总结了dECM水凝胶生物墨水的制备及其基本打印参数;回顾了衍生自不同器官的dECM水凝胶生物墨水的应用,并概括了用于评估这些生物墨水质量的表征方法;得到了制作优质的生物墨水取决于衍生的组织器官、动物和脱细胞方法的选择这一结论。最后,探讨了dECM水凝胶生物墨水的潜在大规模应用以及制造此类生物材料的挑战。
The extracellular matrix(ECM)contains complex protein environments and growth factors that provide mechanical,biophysical,and biochemical signals to cells with tissue-organ specificity.Organderived decellularized ECM(dECM)is currently the most bionic biomimetic material,making it a popular material in 3-D bioprinting applications.This paper summarizes the preparation of dECM hydrogel bio-ink and its basic printing parameters;reviews the application of dECM hydrogel bio-ink derived from different organs,and summarizes the characterization methods used to evaluate the quality of these bio-inks.High quality bio-ink depends on the choice of derived tissue,animal and decellularization methods.Finally,the potential large-scale application of dECM hydrogel bio-inks and the challenges of making such biomaterials are discussed.
