摘要
建立一种用于测定连翘茎中木脂素类成分连翘苷、牛蒡苷元和连翘脂素含量的HPLC方法,并探索不同月份连翘茎中各成分的含量变化规律,为合理开发利用连翘茎资源提供依据。采用Agilent C18柱;流动相为甲醇-0.3%乙酸溶液,流速为0.8 mL·min^-1,检测波长为280 nm,柱温为25 ℃,进样量为10 μL。连翘苷在10~300 μg·mL^-1,牛蒡苷元在2~50 μg·mL^-1,连翘脂素在2~50 μg·mL^-1呈良好的线性关系,平均加样回收率分别为98.55%,97.55%,98.65%,RSD分别为2.96%,2.27%,2.87%。此外,4月和8月采集的连翘茎中连翘苷含量较高,分别为1.68%和0.72%。本研究建立的方法稳定性好、重复性较好,可用于连翘茎中连翘苷、牛蒡苷元、连翘脂素的含量测定;另外,连翘茎中含有较丰富的连翘苷成分,因此,可作为连翘苷制备的原料来源。
To develop an HPLC method for content determination of phillyrin,arctigenin and phillygenin in Forsythia suspensa stems,and explore the content variation of the 3 components of the herbs harvested in different months,and provide the scientific basis of reasonable application.The determination was carried out on an Agilent C18 column eluted with methanol and water containing 0.3%acetic acid in a linear gradient elution.The flow rate was 0.8 mL·min^-1,and the detected wavelength was set at 280 nm.The peak areas and the concentrations of the three components had good linear relationship in the range of 10~300 μg·mL^-1 for phillyrin,2~50 μg·mL^-1 for arctigenin,2~50 μg·mL^-1 for phillygenin.The average recoveries were98.55%,97.55%,98.65%respectively,and RSD were2.96%,2.27%,2.87%respectively.What’s more,the content of phillyrin in the stems of Forsythia suspensa collected in April and August was higher,1.68%and 0.72%,respectively.The method was proved to be accurate,steady,repeatable and can be used for the content determination of phillyrin,arctigenin and phillygenin in Forsythia suspensa stems.In addition,the Forsythia suspensa contains a rich amount of phillyrin,so it can be used as a raw material for the preparation of phillyrin.
