摘要 目的:探讨早孕绒毛和蜕膜组织中的干扰素(IFN-γ)是否上调母胎界面中吲哚胺-2,3双加氧酶(IDO)的表达。方法:收集早孕6~9周的妇女行人工流产获取绒毛组织与蜕膜组织,用Western blot方法检测绒毛、蜕膜组织中IFN-γ与IDO蛋白的表达情况;在绒毛和脱膜组织中加入不同浓度的IFN-γ(1、10和100 ng/ml)分别培养48 h和72 h后,观察IDO蛋白在两种组织中的变化。结果:人早孕绒毛和蜕膜组织均有IFN-γ与IDO蛋白的表达且成正相关性(r=0.987 1,0.979 1,P均<0.01);不同浓度的IFN-γ(1、10和100 ng/ml)分别培养48、72 h后,IDO蛋白表达显著升高(P均<0.05),且随着IFN-γ浓度的增高而升高。结论:在正常生理妊娠状态下,母胎界面中IFN-γ与IDO蛋白质表达成正相关,IFN-γ可上调IDO的表达。 Objective:To investigate whether interferon-γ(IFN-γ)upregulate the expression of indole-2-dioxygenase(indoleamine-2,3-dioxygenase,IDO)in the maternal and fetal interfaces of chorionic villi and decidual tissues of early pregnancy.Methods:The chorionic villi and decidua tissues were obtained by artificial abortion and the expression of IFN-γ and IDO in the villi and decidua tissues were detected by Western blot method.The changes of IDO protein in chorionic villi and decidua tissues were observed by adding different concentrations of IFN-γ(1,10 and 100 ng/ml) for 48 and 72 hours,respectively.Results:There was a positive correlation between IFN-γ and IDO protein in human villi and decidua(r=0.987,0.979;P<0.01).After cultured for 48-72 hours with different concentrations of IFN-γ(1,10 and 100 ng/ml) in the cultured villi and decidua of early pregnancy,the expression of IDO protein was significant(P<0.05),and increased with the increase of IFN-γ concentration.Conclusion:The expression of IFN-γ and IDO protein in maternal and fetal interface is positively correlated with that of normal physiological pregnancy,and IFN-γ can up-regulate the expression of IDO.
