广藿香醇通过调控上皮间质转化抑制人胃癌细胞HGC-27的侵袭和转移 被引量:9

摘要 目的:考察广藿香醇对上皮间质转化(epithelial mesenchymal transition,EMT)及人胃癌细胞株HGC-27侵袭、转移的影响。方法:采用不同浓度广藿香醇体外培养HGC-27细胞株,用MTT法考察广藿香醇对HGC-27细胞增殖的影响;用划痕实验和Transwell实验考察广藿香醇对HGC-27细胞侵袭和迁移的影响;Western印迹法考察广藿香醇对EMT相关蛋白E-cadherin、N-cadherin、β-Catenin、Vimentin表达的影响。结果:广藿香醇能够抑制HGC-27细胞增殖,且呈时间和剂量依赖性,24和48 h的IC50分别为38.9和18.9μmol/L。20和40μmol/L广藿香醇能够显著抑制划痕愈合面积,减少HGC-27细胞迁移和侵袭数量(P<0.05)。20和40μmol/L广藿香醇干预48 h,显著上调E-cadherin蛋白表达,减少N-cadherin、β-Catenin、Vimentin蛋白表达(P<0.05)。结论:广藿香醇可能通过抑制人胃癌细胞HGC-27的EMT而抑制其侵袭、转移能力。 Objective:To investigate the effects of patchouli alcohol on epithelial mesenchymal transition,migration and invasion of human gastric cancer cells HGC-27.Methods:Human gastric cancer cells HGC-27 were cultured in vitro,MTT assay was used to investigate the effect of patchouli alcohol on the growth and proliferation of HGC-27 cells.The effect of patchouli alcohol on the migration and invasion of HGC-27 cells was investigated by scratch and Transwell tests.Western-blot was used to investigate the expression levels of EMT-related proteins(E-cadherin,N-cadherin,β-Catenin,Vimentin).Results:MTT results showed that patchouli alcohol inhibited the proliferation of HGC-27 cells in a time-and dose-dependent manner,and the IC was 38.9μmol/L at 24 h and 18.9μmol/L at 48 h,respectively.20 and 40μmol/Lpatchouli alcohol could significantly inhibit the scratch healing size and reduce the number of migration and invasion of HGC-27 cells(P<0.05).Western-blot showed that the expression of E-cadherin was increased and the expression of N-cadherin,β-Catenin and Vimentin were all decreased(P<0.05)following intervension by 20 and 40μmol/L patchouli alcohol for 48 h.Conclusion:Patchouli alcohol could inhibit the EMT through increasing the expression of E-cadherin and decreasing the expression of N-cadherin,β-Catenin and Vimentin,and inhibit the migration and invasion of HGC-27 cells subsequently.
