摘要
目的:基于网络药理学研究红根草化学成分的分子机制,为其药用植物资源的开发利用提供依据。方法:以中药综合数据库为平台,挖掘红根草化学成分、潜在作用靶点和疾病,通过UniProtKB(UniProt数据库)对潜在作用靶点基因名提取后,再用STRING、KOBAS、Cytoscape软件进行蛋白质-蛋白质相互作用,用京都基因与基因组百科全书(KEGG)通路分析和功能富集分析。结果:红根草4种化学成分包括原儿茶酸、4-羟基红根草邻醌、二氢丹参酮I和丹参酮I,201个潜在作用靶点,204种疾病,构建红根草-化学成分-潜在作用蛋白-疾病的网路关系图;在红根草-化学成分-潜在作用蛋白-KEGG通路网络关系图中有111种信号通路、109个潜在作用蛋白;而关键作用蛋白基因-KEGG通路网络关系图中有87种信号通路、12个关键作用蛋白基因,其包括CASP3、CCND1、BCL2、CDKN1A和TP53等关键基因,其中涉及的主要信号通路和疾病与肿瘤、金黄色葡萄球菌感染、乙型肝炎等有关。结论:红根草作为中药具有多成分、多靶点、协同作用的特点,但对其化学成分的药理机制研究认识缺乏深入研究。
Objective:Based on the network pharmacology,the molecular mechanism of the chemical constituents of Salvia prionitis was studied to provide the basis for the exploitation and utilization of medicinal plant resources.Methods:Based on the traditional Chinese medicine integrated database,the chemical constituents,potential targets and diseases of S.prionitis were mined,and the gene names of potential targets were extracted by UniProtKB.The protein-protein interaction KEGG pathway and function enrichment analysis were analyzed by STRINGK,KOBAS and Cytoscape software.Results:Four chemical constituents of S.prionitis,including protocatechuic acid,4-hydroxyrhizoma orthoquinone,dihydrotanshinone I and tanshinone I,201 potential targets and 204 diseases,were used to construct the network diagram of S.prionitis-chemical components-potential protein-disease,111 signaling pathways and 109 potential proteins in the network diagram of S.prionitis-chemical components-potential protein-KEGG pathway.There are 87 signaling pathways and 12 hub genes in KEGG pathway network,including CASP3,CCND1,BCL2,CDKN1A and TP53.The main signaling pathways and diseases involved are related to tumors,Staphylococcus aureus infection and hepatitis B.Conclusion:As a traditional Chinese medicine,S.prionitis has the characteristics of multi-component,multi-target and synergistic action,but it is lack of understanding of the pharmacological mechanism of its chemical constituents.
