猫细小病毒广西株的分离鉴定及动物发病模型的建立 被引量:6

Isolation and identification of felines parvovirus in Guangxi and establishment of a pathology model

摘要 本研究通过利用FK81细胞按常规方法对猫细小病毒进行分离鉴定。选取其中效价最高的分离株以不同剂量经口腔和鼻腔感染猫,通过测定LD50,观察临床症状、白细胞含量变化、组织病理变化等指标,构建动物发病模型。从86份疑似猫瘟粪便样品中成功分离到6株猫细小病毒,其中GX01株TCID50及血凝价均高于其他5株;不同剂量GX01感染猫后均表现出高热、腹泻、呕吐和体重下降等典型临床症状;感染后第3天粪便中检测到病毒;发病后期白细胞严重减少;攻毒第14天测定LD50为1×10-2.2;解剖表现为肠臌气、部分脏器肿大有出血点;组织病理切片观察到十二指肠和肺上皮细胞出现胞浆、胞核侵染、坏死等明显的组织病变,肾出现脂肪变性。本研究成功分离6株广西地区猫细小病毒并建立发病动物模型,为猫细小病毒发病机制和治疗药物的研究奠定了基础。 In this study,feline parvovirus were isolated and identified using FK81 cells by conventional methods.The isolates with the highest titer were selected to infect cats in oral and nasal cavity at different doses. The LD50 was measured for observations of clinical symptoms,changes of white blood cell content and histopathological changes.From 86 suspected panleukopenia stool samples,six kinds of feline parvoviruses were successfully isolated. The TCID50 and hemagglutination prices of Strain GX01 were higher than that of the other five.Cats infected with different doses of GX01 showed typical clinical symptoms such as high fever,diarrhea,vomiting and weight decline.Viruses were detected in feces on the third day after infection,and leukocytes were severely reduced in the late stage of the disease while the value of LD50 was 1×10-2.2 after 14 days of challenge.Anatomical manifestations were intestinal suffocation and partial organ swelling with bleeding points.Tissue pathological slices displayed cytoplasm,nucleus infection and necrosis in duodenal and lung epithelial cells,and the kidney showed fatty degeneration. In this study,we successfully separated 6 feline parvoviruses from samples collected in Guangxi province and established an animal model for pathology.The results provided a basis for research on the pathogenesis of feline parvovirus and the development of therapeutic drugs.
