参麻益智方对酒精性痴呆大鼠认知功能及海马氧化应激水平的影响 被引量:2

Effects of Shenma Yizhi Decoction on the Cognitive Function and Hippocampal Oxidative Stress in Rats with Alcohol-associated Dementia

摘要 目的探讨参麻益智方对酒精性痴呆大鼠认知功能及氧化应激水平的影响。方法以30只Wistar大鼠为研究对象,随机分为空白组、模型组、中药低剂量组(3.3 g/kg)、中药中剂量组(6.6 g/kg)、中药高剂量组(16.5 g/kg),各6只,分别灌胃蒸馏水及相应剂量中药,3个月后模型组、中药组分别灌胃25%酒精5 g/(kg·d),连续灌胃10 d,空白组仍灌胃等量蒸馏水。观察大鼠行为学变化和海马CA1区椎体细胞形态结构,同时比色法检测大鼠海马组织还原型谷胱甘肽(GSH)、丙二醛(MDA)含量及超氧化物歧化酶(SOD)活力的变化,免疫印迹法检测Nrf2、血红素加氧酶-1(HO-1)蛋白表达。结果中药高剂量组潜伏期较模型组显著缩短(P<0.05),穿台次数及目标象限停留时间百分比较模型组均上升(P<0.05),海马CA1区形态较模型组趋于正常,海马组织中Nrf2及HO-1蛋白表达、GSH含量、SOD活力较模型组均显著升高(P<0.05),MDA含量显著降低(P<0.05)。结论参麻益智方能通过抗氧化应激进而保护神经细胞,改善慢性酒精中毒大鼠的学习记忆功能。 Objective To evaluate the effects of Shenma Yizhi decoction(SMYZD)on the cognitive function and hippocampal oxidative stress in rats with alcohol-associated dementia(AAD).Methods Thirty male wistar rats were randomly divided into five groups:control group,model group,SMYZD low dose group(SMYZD-L),MYZD middle dose group(SMYZD-M),and MYZD high dose group(SMYZD-H).The rats were given distilled water in control group.Model groups,wer giver dose of SMYZD in SMYZD-L,SMYZD-M,and SMYZD-H,respectively.After 3 months,the rats in model group and SMYZD groups were given intragastric administration of 25%alcohol 5 g/kg for 10 days.The rats in control group were administrated with the same amount of distilled water.Then Morrris water maze(MWM)was applied to assess the cognitive function.Morphological alternations in hippocampal CA1 area were observed by Hematoxylin-Eosin(HE)staining.the content of glutathione(GSH)and malondialdehyde(MDA),and the activity of superoxide dismutase(SOD)in hippocampus were measured.The expressions of nuclear factor erythroid 2-related factor(Nrf2)and heme oxygenase-1(HO-1)in the brain tissue were detected by western blot.Results Compared with model group,the escape latency was significantly reduced,the platform cross-over times and the percentage of time spending in the target quadrant swimming were increased in the SMYZD-H group(P<0.05).The morphological deficits of the pyramidal neurons in the CA1 area were alleviated in the SMYZD-H group.The Nrf2 and HO-1 expression,the content of GSH and the activity of SOD significantly increased,while the content of MDA reduced in the SMYZD-H group(P<0.05).Conclusion SMYZD exerts neuro-protective effect by removing oxidations,thus improving the learning and memory ability of rats with alcohol-associated dementia.
