孕酮对缺氧缺血性脑病新生鼠脑组织中超氧化物歧化酶活性和丙二醛含量的影响 被引量:16

摘要 目的探讨孕酮对缺氧缺血性脑病(HIE)新生鼠脑组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量的影响。方法40只7日龄新生Wistar大鼠随机分成5组。假手术组、缺氧缺血组、4 mg孕酮组、8 mg孕酮组和16 m孕酮组。动物先行左侧颈总动脉结扎术,再吸入8%O2和92%N2的混合气体2.5 h建立HIE动物模型。药物组于建立模型前30 min给予不同剂量孕酮溶液,24h后动物全部处死,观察各组新生鼠脑组织中SOD活性和MDA含量的变化。结果缺氧缺血组新生鼠脑组织中SOD活性明显低于假手术组(P<0.01),丙二醛含量明显高于假手术组(P<0.01),药物组SOD活性明显高于缺氧缺血组(P<0.05),丙二醛含量低于缺氧缺血组(P<0.05)。结论孕酮通过升高新生鼠缺氧缺血时脑组织中SOD活性和降低丙二醛含量发挥对缺氧缺血性脑损伤的保护作用。 Objective To investigate the effect of progesterone(PROG) on the SOD activity and the MDA content in brain tis,sue of hypoxic - ischemic encepholopathy (HIE)in newborn rats. Methods Forty 7-day-old neonatal rats were randomly divided into five groups : sham- operated group, hyloxic -ischemic group and 3 pretreatment groups(4 mg,8 mg , 16 mg progesterone respectively) . The rats were subjected to left eorpanon carotid artery ligation, then were exposed to 8% O2 and 92 % N2 for 2.5 h to establish HIE model. The PROG was injected intraperitoneally into the pretreatment groups rats respectively at 30 min before hypoxia. All rats were killed at the 24 h after operations, then the SOD activity and the MDA contents were mensurated. Results The SOD activity in hypoxic ischemic group was significantly lower than that in the shamoperated group(P 〈 0.01 ), the MDA contents was significantly higher than that in the sham - operated group( P 〈 0.01 ). Compared with the hypoxic - ischemic group, the SOD activity in pre treatment groups was significantly higher( P 〈 0.05 ), and the MDA contents in the pre treatment groups was significantly lower ( P 〈 0.05 ) . Conclusion PROK; can increase SOD activity and decrease MDA contents in brain tissue of newborn HIE rats,so it plays an important role in preventing brain against hypoxie- ischemic brain damage by inhibiting the release of free radicals.
