microRNA在多发性骨髓瘤的诊断、预后和靶向治疗中的研究进展 被引量:1

摘要 细胞分子遗传学尤其是microRNA在疾病发生发展过程中有着重要作用.在不同疾病中miRNA具有特定的表达谱,它可精确调控细胞内靶基因和多种信号通路表达,维持机体生理稳态.多发性骨髓瘤(multiplemyeG loma,MM)患者有一系列异常表达的miRNA,其表达水平与肿瘤细胞增殖、染色体畸变、恶性转化有关.且miRG NA表达水平对MM早期的鉴别及监测疾病复发,精准靶向治疗,降低耐药性等均有指导意义.miRNA结合RNA干扰技术有望提高其在MM靶向治疗中的效率及诊断精确度,构建以miRNA为基础的细胞分子遗传学与传统预后分期系统相结合的预后评估体系有望成为未来热点. Molecular genetics,specifically microRNA(miRNA),play an important role in the occurrence and development of diseases.There are specific miRNA expression profiles in different diseases.The miRNA precisely regulate the expression of various intracellular target genes and signal pathways and maintain the physiological homeostasis.Patients with multiple myeloma(MM)have abnormal expression of miRNA,which is associated with tumor cell proliferation,chromosome aberration and malignant transformation.Furthermore,the expression of miRNA is of great significance in early identification,disease recurrence monitoring,precise targeted therapy and reducing drug resistance in MM patients.The miRNA-targeted RNA interference technology is expected to improve the efficiency and diagnostic accuracy in targeted therapy of MM.The construction of prognostic evaluation system based on the combination of miRNA-based molecular genetics and traditional prognostic staging system is expected to become a hot topic in the future.
