一种复合微生态制剂发酵条件优化及其对刺参养殖水体水质的影响 被引量:3

摘要 用实验室保存的6株益生菌,包括3株乳酸菌、2株芽孢杆菌、1株酵母菌进行混合发酵制备成复合微生态制剂。通过优化混合发酵培养基配方及培养条件,确定了6株益生菌混合培养的最佳发酵条件,并对添加不同体积分数(1、5、10 mL/m^3)的复合微生态制剂对刺参养殖水体水质、水体菌群的影响进行了研究。结果表明,6种菌混合培养的最佳培养基配方为葡萄糖15 g/L,可溶性淀粉10 g/L,蛋白胨5 g/L,牛肉膏10 g/L,乙酸钠5 g/L,柠檬酸三铵2 g/L,K2HPO40.15 g/L,MgSO4 0.2 g/L,MnSO4 0.05 g/L,蒸馏水1 L。接种量9%(每株菌等比例混合),加液量为250 mL锥形瓶装液100 mL,温度25℃,初始pH 7.0,摇床转速200 r/min。得到的活菌数为3.54×10^9 cfu/mL,是优化前的2倍。养殖实验结果表明,该复合微生态制剂对水体中硫化物的清除作用不显著,而对氨氮、化学需氧量的清除能力较强,清除率分别达到40.1%和25.0%,最适投菌量为5 mL/m3。水体菌群的测定结果表明,添加复合微生态制剂后,水体中具有净化水体作用的菌群增多,表明该复合微生态制剂对养殖水体菌群具有改善作用。 Six strains of probiotics including three strains of Lactic acid bacteria, two strains of Bacillus and a strain of Saccharomyces were selected and mixed to prepare the compound probiotics. The mixed fermentation medium and culture conditions were optimized, and the formula and parameters of compound probiotics were determined. The effects of the probiotics with different doses of 1, 5, 10 mL/m^3 on the water quality were determined. The results showed that the best mixed culture medium formula is as follows: glucose 15 g/L, soluble starch 10 g/L, peptone 5 g/L, beef paste 10 g/L, sodium acetate 5 g/L, trisodium citrate ammonium 2 g/L, K2HPO4 0.15 g/L, MgSO4 0.2 g/L, MnSO4 0.05 g/L, distilled water 1 L. The other conditions are as follows: inoculation amount of 9 %(equal proportion of each strain), liquid amount of 100 mL, temperature of 25 ℃, initial pH of 7.0, shaking speed of 200 r/min. Under the optimum conditions, the number of viable bacteria could reach to 3.54×10^9 cfu/mL, twice of that before optimization. The results of culture experiment show that the compound probiotics have no significant removal effect on sulfide in water, but have strong removal ability on ammonia nitrogen and COD, reaching 40.1% and 25.0%, respectively, and the optimum dosage is 5 mL/m3 water. The result of water bacterial community shows that the number of bacteria which has the function of purifying the water increased after adding the compound probiotics, indicating that the compound probiotics have the function of improving the culture water.
