Determination of 6 Water-Soluble Vitamins in Polygoni Multiflori Radix from Different Habitants by UPLC-MS/MS

摘要 目的:建立超高效液相色谱-串联三重四极杆质谱联用技术(UPLC-MS/MS)测定何首乌中水溶性维生素类成分的分析方法。方法:将粉碎后的何首乌样品用超纯水超声提取后,经ACQUITY UPLC BEH C18色谱柱(2.1 mm×100 mm,1.8μm)分离并在动态多反应监测模式(MRM)下检测,得到相应的提取离子流图,以峰面积进行定量。结果:何首乌中6个水溶性维生素的含量差别较大,其中VB4为何首乌中维生素的主要来源,在6个维生素类化合物中的含量最高,达到85.89μg/g;VB6平均含量最低,仅为0.68μg/g。其余4种维生素类成分VB2、VB5、NA和NAM的平均含量均相对较低。结论:该法简单快速,结果准确、可靠,重复性好,为何首乌中水溶性维生素类成分的深入研究以及何首乌保健产品的开发提供了参考依据。 Objective:To establish an Ultra-High Performance Liquid Chromatography-Mass/Mass Spectrometry(UPLC-MS/MS)method to determine the contents of water-soluble vitamins in Polygoni Multiflori Radix from different habitants.Methods:The pulverized Polygoni Multiflori Radix sample was ultrasonically extracted with ultrapure water,separated by an ACQUITY UPLC BEH C18 column(2.1 mm×100 mm,1.8μm)and detected under dynamic multiple reaction monitoring mode(MRM)to obtain the corresponding extracted ions Flow chart,and quantified by peak area.Results:The contents of 6 water-soluble vitamins in Polygoni Multiflori Radix had great differences.Among them,the content of VB4 was the highest which was 85.89μg/g;and the content of VB6 was the lowest which was only 0.68μg/g.The average content of the other 4 vitamins,VB2,VB5,NA and NAM,was relatively low.Conclusion:The method is simple,accurate and reproducible.It provides a reference for the follow-up research of water-soluble vatamins and health food development of Polygoni Multiflori Radix.
