重庆云阳橘红褐斑病病原鉴定及生物学特性初步研究

摘要 目的:鉴定从重庆云阳橘红基地采集到疑似橘红褐斑病样品病原菌。方法:使用组织分离、形态学鉴定、真菌ITS序列分析及致病性测定。结果:将其鉴定为橘红褐斑病,病原菌为交链格胞菌(Alternaria alternata)。生物学特性分析表明,该病原菌菌丝生长适宜温度为15~30℃,最适温度为20~25℃,37℃时不能生长;pH值为4~12时均可生长,最适pH值为10;光照对菌丝生长影响不大;该菌可利用多种碳、氮源,最适碳源为果糖和D-甘露醇,最适氮源为蛋白胨和硝酸钠。结论:不同褐斑病菌株间存在一定的差异。 Objective:To identify the pathogens suspected of orange-red brown spot samples collected from the orange-red base in Yunyang,Chongqing.Methods:Tissue isolation,morphological identification,sequence analysis of fungal ITS and pathogenicity were used.Results:It was identified as orange-red brown spot,and the pathogen was Alternaria alternata.The analysis of biological characteristics showed that the suitable temperature for the growth of the pathogenic mycelia was 15~30℃,the optimal temperature was 20~25℃,and it cannot grow at 37℃;it can grow at pH 4~12,and the optimal pH value was 10;Light had little effect on the growth of mycelium;the bacteria can use a variety of carbon and nitrogen sources,the most suitable carbon sources were fructose and D-mannitol,and the most suitable nitrogen sources were peptone and sodium nitrate.Conclusion:There are some differences between different brown spot strains.
