芒果苷通过抑制糖酵解反应影响胃癌细胞迁移及侵袭 被引量:5

摘要 目的:考察芒果苷对胃癌细胞增殖、迁移及侵袭的影响及其与细胞糖酵解反应的关系。方法:考察不同浓度(10,30,90μmol·L^-1)芒果苷对人胃癌SGC-7901和BGC-823细胞增殖、迁移及侵袭的影响,并检测肿瘤细胞的乳酸及葡萄糖消耗水平,同时将芒果苷与丙酮酸激酶M2(PKM2)抑制剂联合(30μmol·L^-1+30μmol·L^-1)给予不同胃癌细胞,初步考察芒果苷影响胃癌细胞迁移及侵袭的机制与胃癌糖酵解反应的关系。结果:芒果苷可浓度依赖性抑制不同胃癌细胞的增殖活力,降低肿瘤细胞迁移及侵袭的能力,并减少细胞乳酸及葡萄糖消耗水平,抑制糖酵解反应,抑制PKM2二聚体的表达,与空白对照组比较差异有统计学意义(P<0.05)。与PKM2抑制剂联合使用后,芒果苷联合抑制剂组与单一抑制剂组相较,对肿瘤细胞增殖活力、细胞迁移及侵袭的的调节作用差异无统计学意义(P>0.05)。结论:芒果苷可通过抑制PKM2二聚体的表达,抑制胃癌细胞糖酵解反应,继而抑制胃癌细胞增殖活力并降低其迁移及侵袭能力。 Objective: To investigate the effects of mangiferin on the proliferation,migration and invasion of gastric cancer cells and its relationship with glycolysis. Methods: Human gastric cancer cell lines SGC-7901 and BGC-823 were used to investigate the effects of mangiferin at different concentrations( 10,30 and 90 μmol · L^-1) on the proliferation,migration and invasion of gastric cancer cells,and the levels of lactic acid and glucose consumption were detected. The effects of mangiferin on glycolysis and the regulation of pyruvate kinase M2( PKM2) protein were clarified. At the same time,mangiferin and PKM2 inhibitors( 30 μmol·L^-1+ 30μmol·L^-1) were given to gastric cancer cells to investigate the effects of mangiferin on the migration and invasion of gastric cancer cells. Results: Mangiferin could inhibit the proliferation of gastric cancer cells in a concentration-dependent manner,reduce the migration and invasion of cancer cells,reduce the consumption of lactic acid and glucose,inhibit glycolysis reaction,inhibit the expression of PKM2 tetramer,and compared with the blank control group,there were significant differences( P < 0.05). When combined with PKM2 inhibitors,there were no significant differences in the regulation of proliferation,cell migration and invasion between mangiferin combined with inhibitors group and the single inhibitor group( P>0.05). Conclusion: Mangiferin can inhibit the proliferation,migration and invasion of gastric cancer cells by inhibiting the expressions of PKM2 and glycolysis of gastric cancer cells.
