摘要
目的探讨STAT4 rs10181656基因多态性与广西地区人群抗中性粒细胞胞浆抗体相关性血管炎(antineutrophil cytoplasmic autoantibodies associated vasculitides,AAV)的易感性及临床症状发生风险的关系。方法聚合酶链式反应(PCR)技术和限制性片段长度多态性(RFLP)方法用于122例AAV患者(病例组)及155例健康人(对照组)基因型的测定,分析基因多态性与AAV易感性关系,并收集临床症状行统计学分析。结果比较病例组和对照组,STAT4 rs10181656基因型和等位基因频率差异均有统计学意义(P<0.05);Logistic回归分析发现,GG基因型频率及G等位基因频率与AAV发病风险显著相关(基因型:OR=2.827,95%CI:1.207~6.621,P=0.017;等位基因:OR=1.537,95%CI:1.066~2.217,P=0.021)。位点基因型及等位基因频率在患者有无血尿亚组中分布差异有统计学意义(P<0.05)。Logistic回归分析发现,GG+CG基因型频率及G等位基因频率与血尿发生风险显著相关(基因型:OR=2.603,95%CI:1.017~2.664,P=0.046;等位基因:OR=2.393,95%CI:1.127~5.081,P=0.023)。结论STAT4 rs10181656基因多态性与广西汉族人群AAV易感性相关,与患者血尿发生相关。
Objective This study attempted to evaluate the association of STAT4 rs10181656 gene polymorphisms with antineutrophil cytoplasmic autoantibodies(ANCAs)associated vasculitides(AAV)and symptoms in Guangxi Han population.Methods The single nucleotide polymorphisms(SNPs)of STAT4 rs10181656 in 277 participants(122 AAVs and 155 healthy controls)were genotyped by PCR-RFLP.The correlation between the genotype and clinical symptoms were evaluated.Results The results showed that both the minor allele frequencies of G allele and GG genotype had significantly differents between AAV patients and controls(P<0.05).Logistic regression analysis found that GG genotype and G allele had significantly relation with the risk of AAV(genotype:OR=2.827,95%CI=1.207~6.621,P=0.017;allele:OR=1.537,95%CI=1.066~2.217,P=0.021).Hematuria were significantly differert in rs10181656 both genotype and allele(P<0.05).Logistic regression analysis found that GG+CG genotype and G allele had significantly relation with the incidence rate of hematuria(genotype:OR=2.603,95%CI=1.017~2.664,P=0.046;allele:OR=2.393,95%CI=1.127~5.081,P=0.023).Conclusion The present study indicates that STAT4 rs10181656 associated with AAV susceptibility in Guangxi Han population,as well as the incidence of hematuria in AAV.
