华蟾素诱导人胶质瘤U87细胞凋亡的作用机制研究 被引量:5

摘要 目的探讨华蟾素(Cinobufacini)对人胶质瘤U87细胞凋亡的促进作用及其机制。方法将体外培养的胶质瘤U87细胞分为对照组、华蟾素组(依华蟾素水平不同分为3个亚组),MTT法测定细胞增殖率,流式细胞仪测定细胞凋亡率,Hoechst 33258核染色法观察细胞核凋亡,透射电镜观察细胞形态学的变化,Western blot法检测胶质瘤U87细胞内Cleaved Caspase-3及Cleaved Caspase-9的蛋白表达水平。结果华蟾素对胶质瘤U87细胞增殖有抑制作用;流式细胞仪及Hoechst 33258核染色法的检测显示,随着华蟾素作用浓度的递增,胶质瘤U87细胞凋亡率呈剂量依赖性递增;华蟾素作用后可使Cleaved Caspase-3及Cleaved Caspase-9蛋白的表达水平升高。结论华蟾素通过改变Cleaved Caspase-3及Cleaved Caspase-9蛋白的表达水平来抑制胶质瘤U87细胞增殖并促进其凋亡。 Objective To study the apoptosis-induced effect of Cinobufacini on glioma U87 cells and its mechanism. Methods In vitro cultured glioma U87 cells were divided into control group and Cinobufacini group(Cinobufacini group were divided into three different subgroup according to the concentrations of glycoside), cell proliferation rate was detected by MTT method, cell apoptosis rate were measured by flow cytometry, nucleus apoptosis was detected by Hoechst 33258 nuclear staining, cell morphological changes were observed by transmission electron microscope, protein expression levels of Cleaved Caspase-3 and Cleaved Caspase-9 were detected by western blot method. Results Cinobufacini had inhibition effect on glioma U87 cells’ proliferation. Flow cytometry and Hoechst 33258 nuclear staining test showed that Glioma cell’s apoptosis rate increased in dose-dependent manner as cinobufacini concentration increasing. Cinobufacini promoted the expression of Cleaved Caspase-3 and Cleaved Caspase-9. Conclusion Cinobufacini inhibited Glioma cell’s proliferation by changing the expression levels of Cleaved Caspase-3 and Cleaved Caspase-9.
