荷叶多酚的提取工艺优化及抗氧化研究 被引量:3

摘要 目的优化荷叶多酚超声波提取法的工艺条件,并研究荷叶多酚的抗氧化性。方法荷叶中多酚的提取采用超声波提取法,通过单因素试验考察乙醇浓度、超声提取时间、料液比和超声提取温度对多酚得率的影响,并利用正交试验优化提取工艺条件。通过邻苯三酚法和Fenton法评价荷叶多酚对超氧阴离子和羟基自由基的清除能力。结果荷叶多酚的最优提取条件为乙醇体积分数60%,提取时间20 min,料液比为1∶12,超声提取温度为50℃。荷叶多酚浓度的为0.25 g/L,其对超氧阴离子的清除率为69.7%,羟基自由基的清除率为66.8%。结论超声波提取荷叶多酚工艺稳定可行,荷叶多酚具有较强的抗氧化性。 Objective To optimize the ultrasonic extraction of polyphenols from lotus leaf and investigate its antioxidant activities.Methods The extraction of polyphenol in lotus leaf was performed by ultrasonic extraction.The effects of ethanol concentration,ultrasonic extraction time,liquid-solid ratio and ultrasonic extraction temperature on the extraction rate of polyphenol were studied by single factor experiments,and orthogonal test was used to optimize the technological conditions.The scavenging ability of polyphenols on superoxide anions and hydroxyl radicals was investigated by the method of pyrolysis and Fenton method.Results The optimal extraction conditions for polyphenol from lotus-leaf as followed:ethanol concentration was 60%,ultrasonic extraction time was 20 min,ratio of liquid to solid was 1∶12,ultrasonic extraction temperature was 40℃.When the concentration of lotus-leaf polyphenols reached 0.25 g/L,the rate of scavenging superoxide anions was 69.7%and hydroxyl radicals was 66.8%.Conclusion The optimum ultrasonic extraction procession is stable and feasible.Lotus leaf polyphenols shows a high antioxidant activity.
