摘要
为了解猪瘟病毒贵州株GZPD E2基因变异情况,为贵州猪瘟疫苗的选择提供参考依据,试验采用RT-PCR方法对GZPD E2基因进行扩增,并对目的基因进行克隆、测序和序列分析。结果:(1)RT-PCR扩增获得大小为1 343 bp的基因片段,与预期扩增大小相符。(2)目的基因克隆、测序分析显示,GZPD与国内毒株shimen株、C株等在 E2基因上存在较高的同源性,为94.9%-99.9%;与国外毒株GPE、Koslov等的核苷酸同源性为95.1%-95.3%。(3)进化分析显示,GZPD与HuBei、C-ZJ-2008、C-strain、HCLV处于同一进化分支,而与shimen、CSFV-GZ-2009、Koslov、GPE处于不同的进化分支。结果表明:猪瘟病毒贵州株GZPD在 E2基因上存在一定变异,提示贵州猪瘟病毒存在一定的进化。
To understand the variation of the GZPD E2 gene of swine fever virus strain in Guizhou province. It provides a reference for the selection of Guizhou swine fever vaccine. The CSFV E2 gene of Guizhou swine fever case was amplified by RT-PCR, and the target gene was cloned, sequenced and sequence analyzed. The results showed that 1 343 bp gene fragment was amplified by RT-PCR, which was consistent with the expected amplification size. The cloning and sequencing analysis of the target gene showed that the CSFV GZPD strain and the domestic strains had high homology of E2 gene, ranging from 94.9% to 99.9%. The nucleotide homology with the foreign strains were 95.1%~95.3%. Evolutionary analysis showed that the CSFV Guizhou strain and HuBei, C-ZJ-2008, C-strain, HCLV strains were in the same evolutionary branch. And in different evolutionary branches with shimen, CSFV-GZ-2009, Koslov, GPE srains. The results showed that the CSFV E2 gene of Guizhou strain had variation, which indicated that CSFV Guizhou strain had evolution, and provided a basis for vaccine selection.
