摘要
目的观察搜风祛痰中药对大鼠心肌缺血再灌注损伤冠脉微循环内皮功能的保护作用并探讨其作用机制。方法健康SPF级Sprague-Dawley大鼠140只,随机分为假手术组、模型组、培哚普利组、搜风祛痰中药组,每组35只。假手术组只穿线不结扎,其余3组建立缺血再灌注模型。预先给予相应药物干预后造模,心电图评价造模情况。ELISA法检测血清内皮素-1(ET-1)、一氧化氮(NO)含量;Western blotting法检测蛋白激酶B(Akt)、磷酸化蛋白激酶B(P-Akt)、内皮型一氧化氮合酶(eNOS)蛋白表达水平。结果心电图显示造模成功。与模型组相比,搜风祛痰中药可升高NO含量、降低ET-1含量,增加P-Akt、eNOS蛋白表达(P<0.05)。结论搜风祛痰中药可保护大鼠心肌缺血再灌注冠脉微循环内皮功能,其机制可能与降低ET-1含量、增加NO含量、上调P-Akt、eNOS蛋白表达相关。
Objective:To observe the protective effect of Soufeng Qutan herbs on the endothelial function of coronary microcirculation in rats with myocardial ischemia-reperfusion injury. Methods:140 healthy SPF SD rats were randomly divided into sham operation group,the model group,perindopril group and TCM group,35 rats in each. The sham operation group was not ligated,and the other groups were ligated to establish ischemia-reperfusion models. The model was established after the corresponding drug intervention and the electrocardiogram was used to evaluate the models. The levels of NO and ET-1 were detected by ELISA;the expression levels of Akt,PAkt and eNOS proteins were measured by Western blotting. Results:The electrocardiogram showed succession for models. Compared with the model group,the levels of NO increased;the levels of ET-1 decreased and the expression of P-Akt and eNOS proteins increased(P < 0.05). Conclusion:Soufeng Qutan herbs can protect the endothelial function of coronary microcirculation in rats with myocardial ischemia reperfusion. Its mechanism may be related to reducing levels of ET-1,increasing levels of NO,and the up-regulation of P-Akt and eNOS proteins.
