摘要
探索珠蓼散对腹泻小鼠模型的治疗作用机理。将50只昆明小鼠随机分成5组,Ⅰ~Ⅲ组为珠蓼散低剂量、中剂量、高剂量试验组,Ⅳ组为常规药物对照组,Ⅴ组为空白对照组。各组均用番泻叶造腹泻模型,Ⅰ~Ⅲ组分别用低剂量、中剂量、高剂量的珠蓼散,Ⅳ组用蒙脱石散灌胃,分别治疗3 d,空白对照组则用生理盐水灌胃3 d,分别于造模后第1、2、3、5、7天采血,检测血清中白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α)的含量,并取空肠进行组织切片观察。结果显示,Ⅰ~Ⅲ组小鼠血清中IL-6与TNF-α的含量均显著低于Ⅴ组(P<0.05),Ⅲ组与Ⅴ组比较,IL-6含量降低28.9%,TNF-α含量降低61.3%;同时Ⅲ组小鼠空肠绒毛平均长度显著高于Ⅴ组,空肠黏膜组织结构修复最快,病理评分最低,平均为2.24。提示珠蓼散对腹泻小鼠的治疗作用机理可能是通过降低炎性因子的分泌和促进受损肠黏膜的修复来实现的。
In order to explore the the rapeutic mechanism of Zhuliao powder on the diarrhea mouse model,50 mice were randomly divided into 5 groups,groupⅠ~Ⅲwere the trial groups with low,medium and high dose of Zhuliao powder,groupⅣwas the conventional medicine control group,while groupⅤwas the control group.The mice in each group were fed with sennaleaf to make diarrhea models.Mice in groupⅠ-Ⅲwere treated with Zhuliao powder in low,medium and high doses for 3 d,groupⅣwith montmorillonite for 3 d,and groupⅤwith saline for 3 d.Blood samples were collected on the 1st,2nd,3rd,5th and 7th day after making models to detect interleukin 6(IL-6)and tumor necrosis factorα(TNF-α)in serum;and the jejunum was taken for tissue section observation.The results showed that the levels of IL-6 and TNF-αin serum of groupⅠ~Ⅲwere significantly lower than those of groupⅤ(P<0.05).Compared with groupⅤ,IL-6 content in groupⅢdecreased by 28.9%,and TNF-αcontent decreased by 61.3%,meanwhile,the average lengths of jejunum villi in groupⅢwas significantly higher than that in groupⅤ;the jejunal mucosae were repaired with the biggest speed,and the pathological score was the lowest,with an average of 2.24.The results showed that the the rapeutic mechanism of Zhuliao powder in diarrhea mice may be achieved by reducing the secretion of inflammatory factors and promoting the damage repair of intestinal mucosa.
