miR-17及其靶基因HIF-1α、STAT3在心源性猝死心肌组织中的表达及其意义 被引量:5

The expression of miR-17, HIF-1α and STAT3 and their correlation in myocardial tissue of cardiac sudden death

摘要 目的观察微小R N A 17(m iR-17)与HIF-lcu STAT3在心脏性猝死(S C D)心肌组织中的表达水平,并探讨其意义。方法选择因心血管系统疾病猝死的20例心脏标本作为S C D组;选取20例死因为非心脏疾患的心脏标本作为对照组。应用免疫组织化学染色方法分别检测两组心肌组织中HlF-lcc和STAT3表达,采用荧光定量PCR方法检测两组心肌组中miR-17、H IF-lot和STAT3的表达。结果免疫组化染色结果显示,两组心肌组织中HIF-lou STAT3的表达比较,差异均有统计学意义(Z=—3.636、一3.552,P<0.01);两组心肌组织中H IF-la与STAT3的表达水平呈负相关(r=-0.996,尸<0.05)。荧光定量P C R方法检测结果显示,S C D组心肌组织中m iR-17与H IF-la相对表达量呈正相关(r=0.706,尸<0.05),m iR-17与STAT3相对表达量呈负相关0=—0.730,尸<0.05)。结论S C D心肌组织中miR-17、H IF-la高表达,STAT3低表达,miR-17、H IF-la和STAT3联合应用有望作为诊断S C D较为客观的指标。 Objective To observe the expression levels of miR-17,H IF-la and STAT3 in myocardial tissue of suddencardiac death (SCD) and explore its significance. Methods Twenty cases of cardiac specimens with sudden death due tocardiovascular diseases were selected as SCD group. 20 cardiac specimens with no heart disease were selected as controlgroup. The expression levels of HIF-laand STAT3 were determined by immunohistochemistry. The expressions of miR-17,HIF-la and STAT3 were detected by real-time PCR. Results The results of immunohistochemical staining showed thatthe expressions of HIF-laand STAT3 in myocardial tissue were statistically significant (Z=-3.636、-3.552, P < 0.01). Theresults of immunohistochemical staining showed that there was a negative correlation between HIF-laand STAT3 expressionin myocardial tissue (r=-〇.996, P < 0.05). The results of real-time PCR showed that there was a positive correlation betweenmiR-17 and H IF-la expression in the SCD group (/*=0.706, P < 0.05), and a negative correlation between miR-17 and STAT3 expression (r= -〇.730, P < 0.05). Conclusion Theexpression of miR-17 and H IF-la in SCD myocardium ishigh, and the expression of STAT3 is low. The combinationof miR-17, H IF -la and STAT3 is expected to be anobjective indicator for the diagnosis of SCD.
