黄芪对哮喘小鼠支气管肺泡灌洗液中IL-2、IL-4及其行为学变化的影响 被引量:6

摘要 目的探讨中药黄芪在哮喘治疗中的作用,观察支气管肺泡灌洗液(BALF)中IL-2、IL-4的变化,阐述黄芪对哮喘的作用机制及治疗效果。方法50只质量18~20g的BALB/c小鼠,周龄在6~8周之间,随机分为5组,每组10只,分别为对照组、哮喘模型组、单纯激素干预组、单纯黄芪干预组以及黄芪+激素干预组,实验组从第1 d至21 d为致敏阶段,均给予卵白蛋白(OVA)腹腔注射,同时黄芪干预组从第1 d起每天给予黄芪液灌胃,激素干预组从第1天给予腹腔注射地塞米松。于实验第22 d分别给予卵白蛋白雾化激发,每天1次,连用7 d。对照组与哮喘模型组小鼠于同一时间给予等量生理盐水灌胃。观察小鼠行为学变化并用酶联免疫吸附试验法检测各组小鼠BALF中IL-2和IL-4含量。结果①激发过程中小鼠行为学变化:黄芪+激素干预组较单纯激素或黄芪干预组与哮喘模型组的小鼠的烦躁不安、呼吸深快、点头运动等症状减轻;②BALF中IL-2、IL-4含量:黄芪干预各组小鼠比较哮喘模型小鼠肺泡灌洗液中IL-2含量升高,IL-4的含量明显减少(P<0.01);黄芪+激素干预组与单纯激素组比较,IL-2含量升高,IL-4减少(P<0.01)。结论黄芪通过调节IL-2,IL-4的含量,改变辅助性T淋巴细胞1与2(Th1/Th2)的比值,缓解哮喘症状,进而起到预防和治疗哮喘的作用。 Objective:Objective:To explore the role of Astragalus membranaceus in the treatment of asthma,observe the changes of IL-2 and IL-4 in bronchoalveolar lavage fluid(BALF),and elaborate the mechanism and therapeutic effect of Astragalus membranaceus on asthma.Methods:Fifty BALB/c mice with a mass of 18~20 g were randomly divided into five groups at the age of 6-8 weeks.Each group consisted of 10 mice.They were divided into control group,asthma model group,hormone intervention group,Astragalus intervention group and Astragalus+hormone intervention group.The experimental group was sensitized from day 1 to day 21,and was intraperitoneally injected with ovalbumin(OVA).At the same time,the Astragalus intervention group was given Astragalus solution daily from the first day,and the hormone intervention group was given dexamethasone intraperitoneally from the first day.On the 22nd day of the experiment,ovalbumin atomization was given once a day for 7 days.The mice in the control group and asthma model group were given the same amount of saline at the same time.The behavioral changes of mice were observed and the contents of IL-2 and IL-4 in BALF of mice in each group were detected by ELISA.Results:1.Behavioral changes in mice during stimulation:The symptoms of restlessness,deep breathing and nodding movement in asthma model group and asthma model group were alleviated in Astragalus+hormone intervention group compared with those in hormone alone group or Astragalus intervention group.2.IL-2 and IL-4 contents in BALF:IL-2 and IL-4 contents in alveolar lavage fluid of asthma model mice were compared in Astragalus intervention group with asthma model mice.The content of IL-4 decreased significantly with the increase of the content(P<0.01).Compared with the simple hormone group,the content of IL-2 increased and the content of IL-4 decreased in the Astragalus+hormone intervention group(P<0.01).Conclusions:Astragalus membranaceus can alleviate asthma symptoms by adjusting the content of IL-2 and IL-4 and changing the ratio of hel
