龙胆总苷对肝纤维化模型大鼠相关指标的改善作用 被引量:2

摘要 目的探讨龙胆总苷对肝纤维化模型大鼠相关指标的改善作用。方法将60只大鼠随机分为正常对照组(A组,等体积0.9%氯化钠溶液),模型组(B组,等体积0.9%氯化钠溶液),秋水仙碱组(C组,0.25 mg/kg),龙胆总苷高、中、低剂量组(D1组、D2组、D3组,100,60,20 mg/kg),各10只。除A组外,其余各组大鼠均腹腔注射15%四氯化碳(CCl4)油溶液(4 mL/kg),首剂加倍,每周2次,连续8周,建立大鼠肝纤维化模型。建模当天,各组大鼠均开始灌胃给予相应药物,每天1次,连续8周。以全自动生化分析仪测定大鼠血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆红素(TBil)水平,以酶联免疫吸附(ELISA)法测定大鼠血清中转化生长因子β1(TGF-β1)、透明质酸(HA)、Ⅲ型胶原(COLⅢ)水平,测定大鼠肝组织匀浆中羟脯氨酸(Hyp)、超氧化物歧化酶(SOD)、丙二醛(MDA)水平;苏木素-伊红(HE)染色后电镜下观察大鼠肝脏组织病理学及炎性改变程度。结果与A组比较,B组大鼠血清中的ALT,AST,TBil,HA,COLⅢ,TGF-β1水平显著升高,肝组织匀浆中Hyp和MDA水平显著升高,SOD水平显著降低(P<0.05),大鼠肝组织病变明显;与B组比较,C组及D1组、D2组、D3组大鼠血清中AST,ALT,TBil,HA,COLⅢ,TGF-β1水平均明显降低,肝组织匀浆中Hyp和MDA水平显著降低,SOD水平显著升高(P<0.05),肝组织损伤程度明显减轻。结论龙胆总苷对肝纤维化模型大鼠各项指标均有较好的改善作用。 Objective To investigate the effect of gentian total glycosides on the improvement of related indexes in rats with liver fibrosis.Methods Totally 60 SD rats were randomly divided into the normal control group(group A,equal volume of 0.9%sodium chloride solution),model group(group B,equal volume of 0.9%sodium chloride solution),colchicine group(group C,0.25 mg/kg),gentian total glycosides high,medium and low dose groups(group D1,group D2 and group D3,100,60,20 mg/kg),10 rats in each group.Except for group A,the rats in the other groups were intraperitoneally injected with 15%CCl4 oil solution(4 m L/kg),the first dose was doubled,twice a week for 8 consecutive weeks,in order to establish the rat model with liver fibrosis.On the day of modeling,the rats in each group were given corresponding drugs by gavage once a day for 8 weeks.The levels of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBil)of rats were measured by automatic biochemical analyzer,and the levels of serum transforming growth factor-β1(TGF-β1),hyaluronic acid(HA),collagen typeⅢ(ColⅢ)of rats were measured by enzyme-linked immunosorbent assay(ELISA),the levels of hydroxyproline(Hyp),superoxide dismutase(SOD)and malondialdehyde(MDA)in the homogenate of rat’s liver were measured.The histopathology and inflammatory changes of rat’s liver were observed by electron microscope after hematoxylin-eosin(HE)staining.Results Compared with those in group A,the levels of serum ALT,AST,TBil,HA,COLⅢ,TGF-β1 of rats were significantly higher,the levels of Hyp and MDA in liver homogenate were significantly higher,while the levels of SOD in liver homogenate were significantly lower in group B(P<0.05),the pathological changes of liver tissue in group B were obvious.Compared with those in group B,the levels of serum AST,ALT,TBil,HA,COLⅢand TGF-β1 of rats were significantly lower,the levels of Hyp and MDA in liver homogenate were significantly lower,while the level of SOD in liver homogenate was significantly higher in g
