马铃薯延薯9号茎尖脱毒及成苗分析 被引量:3

摘要 马铃薯传统种植方法易引起病毒积累和品种退化,目前国内外利用茎尖脱毒防止马铃薯退化,保证马铃薯高产、稳产已成为一种趋势。本文探究不同消毒方法、不同浓度激素配比的诱导培养基、不同茎尖大小、不同培养条件对茎尖成苗及脱毒对马铃薯品种延薯9号的影响。结果表明:自来水冲洗1~2 min+75%乙醇1 min+0.1%升汞5 min+无菌水冲洗2次,可达到理想的消毒效果;诱导培养基MS+1.0 mg·L-1 IAA+1.0 mg·L-1 GA3+30 g·L-1蔗糖+8 g·L-1琼脂、茎尖分生组织0.3~0.4 mm、光照强度2000 lx条件下,光照时长16 h·d-1,培养温度26℃时成苗效果最好。 Traditional potato planting methods are easy to cause virus accumulation and variety degradation.At present,it has become a trend to use virus-free stem tip to prevent potato degradation and ensure high and stable yield.In this paper,the effects of different disinfection methods,different concentrations of hormone ratio of induction medium,different stem tip size,different culture conditions on shoot tip seedling and virus-free on the potato variety Yanshu 9 were studied.The results showed that under the conditions of tap water washing for 1-2 min+75% ethanol for 1 min+0.1% mercuric chloride for 5 min+sterile water washing for 2 times can achieve the ideal disinfection effect.Under the condition of MS+1.0 mg·L-1 IAA+1.0 mg·L-1 GA3+30 g·L-1 sucrose+8 g·L-1 agar,0.3-0.4 mm meristem,2 000 lx light intensity,16 h·d-1 light duration and 26℃culture temperature,the best seedling forming effect was obtained.
