摘要
化疗是肿瘤临床治疗的主要手段,但在治疗过程中,肿瘤细胞易对化疗药物产生耐药性。肿瘤细胞耐药性已成为临床治疗的瓶颈。肿瘤耐药机制涉及多个方面,与药物转运相关蛋白、代谢酶系统的异常表达,DNA损伤的异常修复,肿瘤细胞自噬及凋亡异常等因素有关,其中调控细胞氧化应激反应的核转录相关因子2-抗氧化反应元件(Nrf2-ARE)通路在肿瘤耐药性的产生中发挥关键作用。Nrf2-ARE通路与肿瘤耐药的研究对肿瘤耐药性逆转和肿瘤的治疗有重要指导作用,是未来抗肿瘤分子靶向药物研究的重要发展方向。
Chemotherapy is the main method of clinical treatment,but in the treatment process,tumor cells tend to develop resistance to chemotherapeutic drugs.Drug resistance has become the bottleneck of tumor clinical treatment.The mechanism of tumor drug resistance involves many aspects,including abnormal expression of drug transport-related proteins and metabolic enzyme systems,abnormal repair of DNA damage,abnormal autophagy and apoptosis of tumor cells,etc.The nuclear factor erythroid-2 related factor 2-antioxidant response element(Nrf2-ARE)pathway,which regulates cell oxidative stress,plays a key role in the development of tumor drug resistance.The study of Nrf2-ARE pathway and tumor drug resistance is of instructive significance in the reversal of the resistance and the treatment,which provides an important direction for the future research on the targeted molecular anti-tumor drugs.
