摘要
目的研究槲皮素(quercetin,Q)对高脂膳食小鼠肝脏甲状腺激素(thyroid hormone,TH)作用的影响。方法32只C57BL/6J雄性小鼠随机分为4组:对照组(Con),对照组+0.05%槲皮素组(CQ),高脂组(HF),高脂+0.05%槲皮素组(HFQ)。17w后测定小鼠的血清TH水平,肝脏的氧化还原状态、1型脱碘酶(DIO1)活性,肝脏组织学形态、脂质水平、脂代谢相关的T3靶基因的表达及TH作用关键蛋白水平。结果与对照组相比,高脂组小鼠肝脏脂肪浸润,甘油三酯(triglyceride,TG)、总胆固醇(total cholesterol,TC)、血清总T4及TSH水平显著升高(P<0.05),槲皮素显著降低上述指标(P<0.05)。四组之间总T3水平无显著性差异(P>0.05)。高脂组DIO1活性、mRNA及蛋白水平、肝脏抗氧化物质(GPx、SOD、T-AOC和GSH/GSSG)水平显著下降(P<0.05),MDA显著升高(P<0.05)。与高脂组相比,槲皮素显著改善肝脏氧化还原状态(P<0.05),提高肝脏DIO1和甲状腺激素受体b(TRb)的蛋白水平(P<0.05),显著下调脂质生成基因(Srebf1、Acc1和Fasn)(P<0.05),显著上调脂肪酸氧化基因Cpt1α和能量代谢基因Cox7c和Atp5c1的表达(P<0.05)。在对照组基础上添加槲皮素并未对上述指标产生显著影响。结论槲皮素可能通过维持肝脏氧化还原状态,调控肝脏DIO1和TRb的表达,进而调节脂质代谢相关T3靶基因表达,改善肝脏脂肪变性。
Objective To study the effects of quercetin(Q)on hepatic thyroid hormone action in mice fed a high-fat diet.Methods Thirty-two male C57BL/6J mice were randomly divided into four groups:control group(Con),Con+0.05%quercetin group(CQ),a high-fat diet group(HF),HF+0.05%quercetin group(HFQ).The mice were fed for 17 weeks and then sacrificed.Serum thyroid hormone levels,hepatic redox status,type 1 deiodinase(DIO1)activity,hepatic histologic performance,lipid profiles,T3-dependent lipid metabolism gene expressions and the expression of proteins those are critical to TH action were determined.Results Compared to Con group,lipid accumulation was shown in HF group.The hepatic TG,TC levels,serum total T4 and TSH levels were significantly increased(P<0.05),which could be decreased by quercetin treatment(P<0.05).No significant change of total T3 level was observed among three groups(P>0.05).HF induced significant decrease in hepatic DIO1 activity,mRNA and protein relative level(P<0.05),as well as decreased GPx,SOD,T-AOC and ratio of GSH/GSSG and elevated MDA level compared to Con group(P<0.05).When compared to HF group,quercetin treatment alleviated hepatic redox status(P<0.05),increased hepatic DIO1 and thyroid hormone receptor b(TRb)protein level,significantly down-regulated the expression of lipogenesis genes(including Srebf1,Acc1 and Fasn)(P<0.05),remarkably up-regulated the expression of fatty acid oxidation genes Cpt1αand energy metabolism genes(Cox7c and Atp5c1)(P<0.05).Conclusion Quercetin might maintain hepatic redox status via regulating DIO1 and TRb expression,and there of control T3-dependent lipid metabolism genes expression to improve hepatic steatosis in high fat fed mice.
