拟南芥原生质体瞬时表达系统检测病原菌蛋白激发子的免疫活性 被引量:2

Detection of the Immune Activity of Pathogenic Bacteria Elicitors Using Arabidopsis Protoplast Transient Expression System

摘要 激发子是一类能激活植物防卫反应的化合物,病原菌的某些分泌蛋白可作为激发子激活植物免疫。目前已有许多不同来源的植物免疫激发子被报道,但仍有大量的病原菌分泌型蛋白激发子尚未被鉴定,其触发的植物免疫信号转导途径尚不明确。本研究通过构建四种病原菌分泌蛋白激发子基因(稻瘟病菌MoHrip1及MoHrip2,大丽轮枝菌PevD1,核盘菌SsCut)瞬时表达载体,利用拟南芥原生质体瞬时表达系统获得上述四种分泌蛋白,鉴定其触发植物免疫的活性,初探其参与的植物免疫信号途径。结果表明,MoHrip1、MoHrip2、PevD1和SsCut可引起叶片中活性氧的爆发和胼胝质的积累;它们在拟南芥原生质体细胞中均能激活PTI信号途径下游报告基因FRK1的表达。证明上述四种病原菌蛋白激发子可能参与了植物PTI免疫反应途径的调控;同时也表明本研究采用的原生质体瞬时表达系统可作为快速筛选和鉴定植物免疫激发子的方法。 Elicitors are group of compounds that can initiate plant defense reaction.Some proteins secreted by pathogenic bacteria can act as elicitors to activate plant immunity.At present,although many different types of plant immune elicitors have been reported,a large number of elicitors secreted by pathogenic bacteria have not been identified yet with their downstream signaling pathways remain unclear.In this study,transient expression vectors of four pathogenic bacteria elicitor genes(Magnaporthe oryzae MoHrip1 and MoHrip2,Verticillium dahliae PevD1 and Sclerotiniasclerotiorum SsCut)were constructed and Arabidopsis thaliana protoplasts transient expression system was used to obtain these four secretory proteins.Next,the activities of four elicitors and the plant immune signaling pathway which may be involved in were studied.The results showed that MoHrip1,MoHrip2,PevD1 and SsCut induced reactive oxygen burst and callose accumulation in leaves and activated the expression of FRK1,a reporter of PTI signaling pathway.These results suggest thatMoHrip1,MoHrip2,PevD1 and SsCut may be involved in the regulation of plant PTI immune signaling pathway.It also confirmed that the protoplast transient expression system used in this study can be applied for rapidly screening and identifying unknown plant immune elicitors.
