敲减BMI1对HeLa细胞增殖、侵袭、迁移及凋亡的影响

摘要 目的探讨敲减BMI1对人宫颈癌HeLa细胞系增殖、迁移及凋亡的影响。方法qRT-PCR方法分别检测宫颈癌及正常宫颈组织中BMI1的表达;用siBMI1及siNC转染人宫颈癌Hela细胞作为实验组及对照组,转染24h后,用CCK8实验、划痕实验、Transwell实验、流式细胞术分别检测敲减BMI1对Hela细胞增殖、侵袭、迁移、凋亡能力的影响。结果qRT-PCR结果显示BMI1在宫颈癌组织中的表达显著高于正常宫颈组织;与对照组相比,CCK8实验结果显示敲减BMI1能够有效抑制宫颈癌HeLa细胞增殖的能力;划痕实验的结果表明敲减BMI1可抑制HeLa细胞的迁移能力;Transwell结果表明:敲减BMI1可抑制HeLa细胞的侵袭能力;流式细胞术结果证明:敲减BMI1可以促进HeLa细胞的凋亡。结论敲减BMI1能够显著抑制人宫颈癌HeLa细胞BMI1的表达,降低了HeLa细胞增殖、迁移、侵袭能力,并且可明确诱导HeLa细胞的凋亡。 Objective To investigate the effect of the knockdown of BMI1 on proliferation,migration and apoptosis of human cervical cancer HeLa cell line.Methods The expression of BMI1 in cervical cancer and normal cervical tissues was detected by qRT-PCR,and the effects of knockdown BMI1 on the proliferation,invasion,migration and apoptosis of Hela cells were detected by CCK8 assay,scratch assay,transwell assay and flow cytometry on 24 hours after siBMI1 and siNC were used as experimental group and control group.Results The results of qRT-PCR showed that the expression of BMI1 in cervical cancer tissues was significantly higher than that in normal cervical tissues.Compared with the control group,CCK8 assay showed that knockdown of BMI1 could effectively inhibit the proliferation of cervical cancer HeLa cells,scratch assay showed that knockdown of BMI1 could inhibit the migration of HeLa cells,Transwell results showed that knockdown BMI1 could inhibit the invasion of HeLa cells.The results of flow cytometry showed that knockdown of BMI1 could promote the apoptosis of HeLa cells.Conclusion Knockdown of BMI1 could significantly inhibit the expression of BMI1 in human cervical cancer HeLa cells,reduce the proliferation,migration and invasion of HeLa cells,and induce apoptosis of HeLa cells.
