摘要 目的探讨儿童急性淋巴细胞白血病(ALL)患者骨髓中miRNA-326的表达及其临床意义。研究N-ras基因在儿童ALL微小残留病(MRD)中的表达,以期为儿童ALL的复发提供生物学检测标志。方法本研究选取35例儿童ALL和35例非白血病儿童作为对照组,通过实时荧光定量PCR(RT-PCR)检测miRNA-326在儿童ALL中相对表达量,使用单链构象多态性PCR(PCR-SSCP)检测N-ras基因在治疗1年后的微小残留病(MRD)中的变化。结果miRNA-326在儿童ALL中的相对表达量(2.22±0.72)较对照组(6.35±1.23)明显下降;SSCP的电泳结果显示MRD组患者N-ras基因的电泳速度较对照组快,基因突变率为44.44%。结论miRNA-326和N-ras基因的检测可作为儿童ALL的诊断及复发生物学标志,也可为白血病的耐药性监测和治疗提供新的靶点。 Objective To investigate the expression and clinical significance of miRNA-326 in bone marrow of pediatric acute lymphoblastic leukemia(ALL).And to study the expression of N-ras gene in ALL minimal residual disease(MRD),which provide biological markers for the recurrence of ALL.Methods A total of 35 children with ALL and 35 children with non-leukemia were selected as control group.Real-time fluorescence quantitative PCR(RT-PCR)was used to detect the relative expression of microRNA-326 in children with ALL,and single strand conformation polymorphism PCR(PCR-SSCP)was used to detect the changes of N-ras gene in MRD after 1 year treatment.Results The relative expression of miRNA-326 in pediatric ALL(2.22+0.72)was significantly lower than that of control group(6.35+1.23).The electrophoresis results of SSCP showed that the electrophoresis speed of N-ras gene in MRD group was faster than that of control group.The mutation rate was 44.44%.Conclusion The detection of miRNA-326 and N-ras genes can be used as biological markers for the diagnosis and recurrence of pediatric ALL,which can also provide new targets for drug resistance monitoring and treatment of leukemia.
