摘要
目的:建立RP-HPLC法同时测定小儿宣肺止咳颗粒中盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷、黄芩苷、升麻素苷、槲皮素-3-O-β-D-葡萄糖-7-O-β-D-龙胆双糖苷、芥子碱硫氰酸盐、甘草酸铵的含量。方法:色谱柱:ZORBAX Eclipse XDB-C18(250 mm×4.6 mm,5μm);流动相:甲醇-0.1%磷酸溶液(含0.04%三乙胺和0.02%二丁胺),梯度洗脱;检测波长:254 nm;流速:1.0 ml·min^-1;柱温:30℃;进样量:10μl。结果:盐酸麻黄碱、盐酸伪麻黄碱、苦杏仁苷、黄芩苷、升麻素苷、槲皮素-3-O-β-D-葡萄糖-7-O-β-D-龙胆双糖苷、芥子碱硫氰酸盐、甘草酸铵检测质量浓度的线性范围分别为6.405~57.64,7.845~70.60,36.96~332.6,78.30~704.7,3.126~28.13,2.844~25.59,3.478~31.30,15.78~142.0μg·ml^-1(r值范围0.9994~0.9999);平均加样回收率分别为98.6%,99.2%,99.0%,99.1%,98.7%,98.9%,98.5%,99.0%(RSD<2.0%,n=6);精密度、重复性、稳定性(48 h)试验的RSD<2.0%(n=6)。结论:该方法简单、有效、结果准确,可用于小儿宣肺止咳颗粒中上述8种成分含量的同时测定。
Objective:To establish an HPLC method for the content determination of 8 constituents in Xiaoer Xuanfei Zhike granules.Methods:The determination was performed on a ZORBAX Eclipse XDB C18 column(250 mm×4.6 mm,5μm)with the mobile phase consisting of methanol-0.1%phosphoric acid(containing 0.04%trimethylamine and 0.02%dibutylamine).The flow rate was 1.0 ml·min^-1 with gradient elution.The detection wavelength was set at 254 nm.The column temperature was set at 30℃and the injection volume was 10μl.Results:The linear range was 6.405-57.64μg·ml^-1 for ephedrine hydrochloride,7.845-70.60μg·ml^-1 for pseudoephedrine hydrochloride,36.96-332.6μg·ml^-1 for amygdalin,78.30-704.7μg·ml^-1 for baicalin,3.126-28.13μg·ml^-1 for prim-O-glucosylcimifugin,2.884-25.59μg·ml^-1 for quercetin-3-O-β-D-glucose-7-O-β-D-gentiobioside,3.478-31.30μg·ml^-1 for sinapine cyanide sulfonate and 15.78-142.0μg·ml^-1 for ammonium glycyrrhizate(r=0.9994-0.9999).The average recovery was 98.6%,99.2%,99.0%,99.1%,98.7%,98.9%,98.5%and 99.0%(RSD<2.0%,n=6),respectively.The RSDs of precision,reproducibility and stability tests(48 h)were lower than 2.0%(n=6).Conclusion:The method is simple,effective and accurate,which can be used for the simultaneous determination of 8 active constituents in Xiaoer Xuanfei Zhike granules.
