近江牡蛎糖胺聚糖降血糖及抗凝血活性研究 被引量:8

Hypolipidemic and anticoagulant activities of glycosaminoglycan from oyster

摘要 以近江牡蛎糖为原料,经酶解制得牡蛎糖胺聚糖粗制品,进行体外降血糖及抗凝血活性研究。根据不同浓度牡蛎糖胺聚糖对α-葡萄糖苷酶活性抑制作用效果,选出抑制作用效果最佳的浓度,测定在不同反应时间的抑制作用效果以及对酶的动力学测定;采用鲫鱼血作为血浆,以体外抗凝血原时间和抗凝血复钙时间来进行牡蛎糖胺聚糖的体外抗凝血评价。结果表明:牡蛎糖胺聚糖对α-葡萄糖苷酶活性具有一定抑制作用,最佳浓度为0.4 mg/mL,抑制率最高达到43.68%;作用时间10min抑制率达到最大,抑制率为66.7%;由动力学试验判断出牡蛎糖胺聚糖对α-葡萄糖苷酶的抑制类型为反竞争性抑制;牡蛎糖胺聚糖浓度为1 mg/mL时,抗凝血原时间与阳性对照肝素钠(0.1 mg/mL)的抗凝血原时间等效,同时使凝血的复钙时间的增加,呈现出非常好的抗凝血效果及性能。 The crude oyster glycosaminoglycan(GAG) was prepared from Jinjiang oyster glycosaminoglycan by enzymatic hydrolysis. The inhibition effect of oyster GAG on α-glucosidase activity by different concentrations was selected to determine the best concentration of inhibition effect. The inhibition effect of oyster GAG on α-glucosidase activity at different reaction times and the kinetics of enzyme were determined. Crucian blood was used as plasma to evaluate the anticoagulant effect of oyster GAG in vitro. The results showed that oyster GAG inhibited the activity of α-glucosidase to some extent. The optimal concentration was 0.4 mg/mL, and the inhibition rate was up to 43.68%. The inhibition rate reached the maximum at 10 min, and the inhibition rate was 66.7%. The inhibition type of oyster GAG to α-glucosidase was determined by kinetic test to be anticompetitive. When the concentration of oyster GAG is 1 mg/mL, the anticoagulant time is equivalent to that of the positive control heparin sodium(0.1 mg/mL), and it can increase the time of coagulation calcium compound, presenting very good anticoagulant effect and performance.
