黑龙江省某规模化奶牛场牛病毒性腹泻病毒qRT-PCR结合双抗夹心ELISA的检测 被引量:3

Detection of Bovine viral diarrhea virus using qRT-PCR combined with double-antibody sandwich ELISA in a large-scale cattle farm in Heilongjiang Province

摘要 为了对黑龙江省规模化奶牛场牛病毒性腹泻/黏膜病进行检疫净化,试验应用qRT-PCR法检测了黑龙江省4个规模化奶牛场(A^D场)奶牛耳组织样品(共1286份,58组),qRT-PCR法检测得到的阳性组样品再用双抗夹心ELISA进行每份样品的检测。阳性牛间隔1个月用RT-PCR方法复检。结果表明:A场牛病毒性腹泻病毒阳性率为0.5%(1/193),B场为0.1%(2/875),C场为1.8%(1/55),D场未检测出牛病毒性腹泻/黏膜病抗原阳性牛(0/163)。间隔1个月,用RT-PCR方法复检,阳性符合率100%。说明qRT-PCR结合双抗夹心ELISA方法适合于规模化奶牛场牛病毒性腹泻/黏膜病的快速、特异性的检疫净化。 To quarantine porification the infection of bovine viral diarrhea-mucosal disease(BVD-MD)in large-scale cattle farms in Heilongjiang Province,ear tissue samples of dairy cattle form(a total of 1286,58 groups)four large-scale dairy farms(named A,B,C,D)in Heilongjiang Province were tested by qRT-PCR and double-antibody sandwich ELISA.The positive cattle were rechecked by PCR one month later.The results showed that the Bovine viral diarrhea virus(BVDV)-positive cow was detected in 193 cattle in the farm A,with a positive rate of 0.5%(1/193);two BVDV positive cattle were detected in the farm B,the positive rate was 0.1%(2/875);one BVDV positive cow was detected in the farm C,the positive rate was 1.8%(1/55);no BVDV antigen-positive cattle were detected in the farm D(0/163).After one month interval,the positive coincidence rate was 100%retested by RT-PCR method.It indicates that qRT-PCR combined with double-antibody sandwich ELISA are rapid and specific and suitable for quarantine and removal of BVDV in large-scale dairy farm.
