摘要
目的探讨载前列腺素E2(prostaglandin E2,PGE2)的乳酸-羟基乙酸共聚物微球(PLGA-PGE2-MSs)对成骨样细胞株MC3T3-E1成骨因子表达的影响。方法采用膜乳化-溶剂挥发法制备粒径均匀的PGE2-PLGA微球。以无添加物培养基(n=6)、空白微球(n=6)为对照组;PGE2(n=6)及缓释PLGA-PGE2-MSs(n=6)为实验组。PGE2浓度为0.0125、0.0500、0.2000、0.8000μmol/L的各实验组与成骨样细胞株MC3T3-E1孵育5、7、9、11 d;采用RT-PCR检测不同时间成骨因子的表达水平。结果微球对照组与空白对照组各时间点比较差异无统计学意义(P>0.05);不同浓度的PGE2、PLGA-PGE2-MSs组与空白对照组同一时间点比较,均能促进RUNX2、ALP、OPN基因表达水平(P<0.05);相同PGE2浓度水平的两个实验组同一时间点比较,PLGA-PGE2-MSs组促进基因表达水平高于PGE2组(P<0.05)。结论PLGA-PGE2-MSs具有明显的缓释作用;在本研究设定的浓度区间内,PGE2、PLGA-PGE2-MSs均可促进MC3T3-E1细胞增殖、分化、基质成熟等不同阶段代表性基因RUNX2、ALP、OPN的表达水平。
Objective To explore the effect of PLGA microspheres loaded with prostaglandin E2(PLGA-PGE2-MSs)on the expression of osteogenic factor from osteoblast-like cell line MC3T3-E1.Methods PGE2-PLGA-MSs with uniform particle size were prepared by membrane emulsification solvent evaporation method.No additive medium(n=6)and blank microsphere(n=6)were used as control group,PGE2(n=6)and sustained releasing PLGA-PGE2-MSs(n=6)were used as experimental group.Each experimental group with PGE2 concentration of 0.0125,0.0500,0.2000,and 0.8000μmol/L was incubated with the osteoblast-like cell line MC3T3-E1 for 5,7,9,and 11 days,and the expression level of osteogenic factor was detected by RT-PCR at different time.Results There was no significant difference between microsphere control group and blank control group at each time point(P>0.05);Compared with the blank control group at the same time point,different concentrations of PGE2 and sustained releasing PLGA-PGE2-MSs could promote the expression of Runx2,ALP and OPN genes(P<0.05).At the same time point,in the two experimental groups with the same PGE2 concentration level,the expression level of gene in the sustained releasing PLGA-PGE2-MSs group was higher than that in the PGE2 group(P<0.05).Conclusions PLGA-PGE2-MSs have obvious sustained releasing effect.Within the concentration range set in this study,PGE2 and PLGA-PGE2-MSs can promote the expression level of Runx2,ALP and OPN in MC3T3-E1 cells at different stages of proliferation,differentiation and matrixmaturation.
