铁死亡机制及在肿瘤中的应用进展 被引量:8

Mechanisms of ferroptosis and its applications in cancers

摘要 铁死亡是一种不同于凋亡、坏死、自噬的调节性细胞死亡形式。其特殊性表现为脂质过氧化物堆积、线粒体体积缩小。铁死亡发生的核心为活性氧的产生及谷胱甘肽依赖性过氧化物酶4的还原反应。有多种诱导剂及抑制剂调控铁死亡过程,在正常组织中铁死亡表现为衰老和退行性改变,而在肿瘤组织中,可通过抑制铁死亡发生而促进肿瘤生长。目前已有药物诱导肿瘤细胞铁死亡发生,与传统治疗方式联合应用有望成为新的治疗手段。 As a regulated cell death,ferroptosis is different from apoptosis,necrosis and autophagy.Its particularity is lipid peroxide accumulation and mitochondrial size reduction.There are many inducers and inhibitors to regulate the ferroptosis process by regulating the production of reactive oxygen species and the reduction of glutathione-dependent peroxidase 4,which play important roles in ferroptosis.In normal tissues,ferroptosis is characterized by aging and degenerative changes,while in cancer tissues,ferroptosis can be inhibited in many ways,thus promoting the growth of tumors.At present,several drugs have been used to promote ferroptosis in cancer cells,and the combination of traditional therapies is expected to become a new treatment mode.
