开郁清热方通过调节胰岛细胞凋亡保护胰岛功能的机制研究 被引量:9

Mechanism Study on Depression-Relieving and Heat-Clearing Recipe Protecting Pancreas Islet Function by Regulating Pancreas Islet Cell Apoptosis

摘要 [目的]探讨中药开郁清热方对胰岛功能的作用及其机制.[方法]以高糖诱导INS-1细胞构建胰岛β细胞功能衰竭模型,分别给予开郁清热方大、小剂量含药血清处理,以罗格列酮含药血清处理作为阳性对照.流式细胞术检测细胞凋亡,逆转录聚合酶链反应(RT-PCR)法检测细胞Sirt1、叉头状转录因子O1(FOXO1)mRNA表达,免疫组化法检测细胞凋亡相关基因Bcl-2、Bax、p53的蛋白表达.[结果]开郁清热方大剂量含药血清可有效降低高糖诱导的INS-1细胞凋亡(P<0.05),升高细胞Sirt1 mRNA、Bcl-2蛋白表达水平,降低FOXO1 mRNA及Bax、p53蛋白表达水平(均P<0.05).[结论]开郁清热方可通过调节胰岛β细胞凋亡起到保护胰岛功能的作用. Objective To explore the effect of Depression-relieving and Heat-clearing Recipe(DHR)on pancreas islet function and its mechanism.Methods INS-1 cells were induced to pancreas isletβ-cell function failure model by high glucose,and then were respectively given treatment with the serum containing big-and smalldosage of DHR,the serum containing rosiglitazone treatment being set up as positive control.The cell apoptosis was measured by flow cytometry,the mRNA expression levels of Sirt1 and forkhead box transcription factor class O1(FOXO1)were determined by reverse transcription polymerase chain reaction(RT-PCR),and the protein expression levels of the cell apoptosis-related genes such as Bcl-2,Bax and p53 were detected by immunohistochemistry.Results DHR was effective for reducing the apoptosis of INS-1 cells induced by high glucose(P<0.05),increasing the mRNA expression level of Sirt1 and protein expression level of Bcl-2,and decreasing the mRNA expression level of FOXO1,and protein expression levels of Bax and p53(P<0.05).Conclusion DHR plays a role in protection of pancreas islet function by regulating the apoptosis of pancreas isletβ-cells.
