从Kiss1、GPR54 mRNA表达水平探讨过度瘦身对大鼠生殖功能的影响及左归丸的干预作用 被引量:4

Exploring Effect of Excessive Weight Loss on Reproductive Function of Rats and Intervention Actions of Zuogui Pills Based on mRNA Expression Levels of Kiss1 and GPR54

摘要 [目的]探讨过度瘦身对大鼠生殖功能的影响及左归丸的干预机制.[方法]选用SPF级健康雌性动情周期规律的12周龄SD大鼠80只,随机分为A、B组,每小组再分为空白组、模型组、恢复饮食组、恢复饮食+左归丸组,每组10只.A组造模4周,B组造模8周,每组恢复4周.采用半量饮食+过度运动+左旋肉碱法复制瘦身大鼠模型.每日记录大鼠一般状态及每周记录1次大鼠体质量变化.分别于4、8、12周后,采用酶联免疫吸附分析(ELISA)检测A、B组大鼠血清性激素促卵泡激素(FSH)、促黄体激素(LH)、雌二醇(E2)含量,荧光定量逆转录聚合酶链反应(qRT-PCR)技术检测大鼠下丘脑人吻素1(Kiss1)及G蛋白偶联受体54(GPR54)mRNA表达水平.[结果](1)与同组空白组比较,模型A、B组大鼠体质量明显下降(P<0.01);与同组模型组比较,恢复饮食A、B组大鼠体质量均明显升高(P<0.01);与同组恢复饮食组比较,恢复饮食+左归丸A组大鼠体质量升高(P<0.05).(2)恢复饮食A组、恢复饮食+左归丸A组动情周期规律的大鼠数量较模型A组升高(P<0.05或P<0.01).(3)与同组空白组比较,模型A、B组FSH、LH水平均明显升高,E2水平均明显下降(P<0.01).与同组模型组比较,恢复饮食组FSH、LH水平均明显下降,E2水平明显升高(P<0.01).与同组恢复饮食组比较,恢复饮食+左归丸A组的FSH、LH明显下降(P<0.01),E2水平升高(P<0.05).A、B组之间对应组别比较,A组FSH、LH水平明显低于B组,E2水平均明显高于B组(P<0.01).(4)模型A、B组大鼠Kiss1、GPR54 mRNA的表达水平较同组空白组显著降低(P<0.01);恢复饮食A组大鼠Kiss1、GPR54 mRNA的表达水平较模型A组显著上升(P<0.01);与恢复饮食A组比较,恢复饮食+左归丸A组大鼠Kiss1、GPR54 mRNA表达显著升高(P<0.01).造模8周各组Kiss1、GPR54 mRNA的表达水平较造模4周对应组显著下降(P<0.01).[结论]①通过"半量饮食+过度运动+左旋肉碱"的方法可以导致大鼠体质量逐渐下� Objective To explore the effect of excessive weight loss on reproductive function of rats and the intervention mechanism of Zuogui Pills.Methods Eighty SPF healthy 12-week female SD rats of regular estrus cycle were randomly divided into group A and B,and the group A and B were further respectively divided into blank group,model group,diet recovery group,diet recovery plus Zuogui Pills group,10 rats in each group.The modeling in group A covered for 4 weeks,group B for 8 weeks,and then each group restored for 4 weeks.A weight loss model was established by combination of half the amount of diet,over exercise and l-carnitine.The general state of the rats was recorded daily and body mass changes in rats were recorded weekly.After 4,8,12 weeks,the serum sex hormones such as folliclc stimulating hormone(FSH),luteinizing hormone(LH),estradiol(E2)contents in group A and B were separately detected by enzyme-linked immunosorbent assay(ELSIA),and the mRNA expression levels of human kissin 1(Kiss1)and G-protein-coupled receptor 54(GPR54)in the hypothalamus of rats were determined by quantitative fluorescence reverse transcription-polymerase chain reaction(qRT-PCR)assay.Results(1)Compared with the intra blank group,the body mass of rats in the model group A and B was decreased significantly(P<0.01).Compared with the intra model group,the body mass of rats in the recovery diet group A and B was significantly increased(P<0.01).Compared with the intra recovery diet group,the body mass of rats in the diet recovery plus Zuogui Pills group A was increased(P<0.05).(2)The number of rats of regular estrus cycle in the recovery diet group A and diet recovery plus Zuogui Pills group A was higher than that in the model group A(P<0.05 or P<0.01).(3)Compared with the intra blank group,FSH and LH levels in the model group A and B were significantly increased,and E2 level was significantly decreased(P<0.01).Compared with the intra model group,FSH and LH levels in the recovery diet group A and B were significantly decreased,and E2 level was s
