摘要 以藜麦苗为原料,采用响应面法对藜麦叶中黄酮的提取工艺进行了优化。首先采用单因素试验,研究了乙醇体积分数、提取时间、提取温度和液料比对藜麦叶黄酮提取量的影响;在此基础上,采用Design-Expert软件的中心组合设计方法设计响应面试验,研究了乙醇体积分数、提取时间、提取温度和液料比各自变量及其交互作用对黄酮提取量的影响,并建立了以黄酮提取量为响应值的二次多项式回归数学模型,据此得到藜麦叶黄酮提取的最佳工艺。结果表明:藜麦叶黄酮的最佳提取工艺条件为乙醇体积分数68%、液料比32∶1、提取时间100 min、提取温度61℃,该条件下黄酮提取量可以达到(12.41±0.03)mg/g,与测定值接近。说明此响应面法得到的回归模型具有一定的可靠性,可为藜麦叶提取黄酮在生产上提供一定的借鉴作用。 The extraction process of flavonoids from quinoa leaves were optimized by response surface methodology.Firstly,the effects of ethanol volume fraction,extraction time,extraction temperature and liquid material ratio on the extraction of flavonoids were studied by single factor experiment.On the basis of these,the interaction effects of these factors were studied by designing response surface test of Design-Expert software.The mathematical model of quadratic polynomial regression was established using the extraction amount of flavonoidsas the response value to obtain the optimum extraction process.The results showed that the optimal extraction process of flavonoids was ethanol volume fraction of 68%,liquid material ratio of 32∶1,extraction time of 100 min,and extraction temperature of 61℃.Under this condition,the extraction amount of flavonoids could reach(12.41±0.03)mg/g,which was close to the test value,indicating that the regression model obtained by this response surface method had certain reliability,which could provide reference for the extraction of flavonoids from quinoa leaves.
