乳康胶囊对乳腺增生大鼠乳腺组织及相关受体表达的影响 被引量:4

Effects of Rukang Capsule (乳康胶囊) on Expressions of Estrogen and Related Receptor in Breast Tissue of Rats with Hyperplasia of Mammary Glands

摘要 目的:观察乳康胶囊对大鼠乳腺增生组织及雌孕激素受体表达的影响。方法:48只雌性健康未孕大鼠,随机抽取8只作为正常对照组,其余40只大鼠采用注射苯甲酸雌二醇及黄体酮建立大鼠乳腺增生模型,将造模成功的大鼠随机分为5组,即模型组,阳性对照组,乳康胶囊小、中、大剂量组。各组大鼠给予相应药物干预后HE染色观察各组大鼠乳腺组织变化,采用酶联免疫吸附(ELISA)法测定大鼠乳腺组织血管内皮生长因子(VEGF)含量,采用免疫组化法测定各组大鼠乳腺组织血管内皮生长因子(VEGF)、乳腺雌激素受体α(ERα)和孕激素受体(PR)表达。结果:与正常对照组相比,模型组大鼠乳腺组织中腺泡数目增多,腺泡和导管增生、扩张,细胞排列紊乱,表现出典型的乳腺增生症状,其乳腺组织VEGF、ERα和PR表达水平升高(P<0.05);与模型组相比,乳康胶囊各剂量组大鼠乳腺增生小叶和腺泡数量减少,同时乳腺组织VEGF、ERα和PR表达降低。结论:乳康胶囊对大鼠乳腺增生具有抑制作用,其治疗机制可能与下调VEGF、ERα和PR的表达有关。 Objective:To observe the effects of Rukang Capsule on the hyperplasia of mammary glands and the expressions of estrogen and progesterone receptors in rats.Methods:Forty-eight SPF-grade female healthy infertile rats were randomly selected as normal control group after one week of adaptive feeding.The other 40 rats were injected with estradiol benzoate and progesterone to establish mammary gland hyperplasia model.Rats were randomly divided into five groups:model group,positive control group and Rukang capsule small,medium and large dose groups,HE staining was used to observe the changes of mammary gland tissues in rats after drug intervention.The content of vascular endothelial growth factor(VEGF)in mammary gland tissues was determined by enzyme-linked immunosorbent assay(ELISA),and the expressions of VEGF,estrogen receptor alpha(ER alpha)and progesterone receptor(PR)were detected by immunohistochemistry.Results:Compared with the normal control group,the number of acini increased,the proliferation and expansion of acini and duct,the disorder of cell arrangement and the typical hyperplasia of mammary gland were found in the model group,and the expression levels of VEGF,ER alpha and PR were increased(P<0.05).The number of proliferating lobules and acinus decreased,the expressions of VEGF,ER and PR in breast tissue were reduced.Conclusion:Rukang Capsule can inhibit the hyperplasia of mammary gland in rats,and its therapeutic mechanism may be related to the down-regulation of the expressions of VEGF,ER alpha and PR.
