高压氧对脑缺血再灌注大鼠血清心型脂肪酸结合蛋白的影响

摘要 目的探讨高压氧对脑缺血再灌注大鼠血清心型脂肪酸结合蛋白的影响。方法选取SD成年雄性大鼠60只,采用随机数表法分为高压氧治疗组、大脑中动脉栓塞组和假手术组,各20只。大脑中动脉栓塞组大鼠行常规心脑肺复苏(CPR)治疗,高压氧组大鼠在CPR治疗后5 h内进入动物高压氧舱内,设置2.5ATA压力按照60 min/次,连续治疗3 d。所有动物均于治疗后第1、4和7 d完成神经功能缺陷评分,并经过免疫组化步骤对每组的大鼠血清心型脂肪酸结合蛋白表达情况进行分析。结果高压氧组大鼠的神经功能缺陷评分良好改善,高压氧前、前后的神经功能缺陷评分分别为(46.81±1.81)m、(20.18±0.99)m,且神经元损伤程度较其他两组明显轻,差异具有统计学意义(P<0.05);高压氧组大鼠的血清心型脂肪酸结合蛋白表达较大脑中动脉栓塞组明显高,差异具有统计学意义(P<0.05)。结论对脑缺血再灌注行高压氧治疗,能够有效保护大脑中动脉缺血再灌注损伤,能有效改善神经功能,这与血清心型脂肪酸结合蛋白表达增高有关。 Objective To investigate the effect of hyperbaric oxygen on serum cardiac fatty acid binding protein in rats with cerebral ischemia reperfusion.Methods 60 adult male SD rats were randomly divided into hyperbaric oxygen treatment group,middle cerebral artery embolization group and sham operation group,20 rats each.Rats in the middle cerebral artery embolism group were treated with conventional cardio-cerebral-pulmonary resuscitation(CPR).Rats in the hyperbaric oxygen group entered the hyperbaric oxygen chamber within 5 hours after CPR treatment.The pressure of 2.5 ATA was set at 60 min/time for 3 days.The neurological deficit scores were completed on the 1st,4th and 7th day after treatment,and the expression of serum cardiac fatty acid binding protein in each group was analyzed by immunohistochemical steps.Results The neurological deficit scores of rats in hyperbaric oxygen group were improved well.The neurological deficit scores before and after hyperbaric oxygen group were(46.81±1.81),(20.18±0.99).The degree of neuronal injury in hyperbaric oxygen group was significantly lighter than that in other two groups(P<0.05).The serum cardiac fat of rats in hyperbaric oxygen group was significantly lower than that in other two groups(P<0.05).The expression of acid binding protein was significantly higher than that of middle cerebral artery embolization group(P<0.05).Conclusion Hyperbaric oxygen therapy for cerebral ischemia-reperfusion can effectively protect middle cerebral artery from ischemia-reperfusion injury and improve neurological function,which may be related to the increased expression of serum cardiac fatty acid binding protein.
