摘要
利用响应面分析方法对螺旋藻细胞的酶解破壁工艺进行优化。以破壁率为指标,通过单因素试验分别考察料液比、加酶量、酶解温度、时间对螺旋藻破壁的影响。采用单因素试验和Box-Behnken中心组合试验原理确定酶解破壁条件,结果表明,螺旋藻细胞破壁的最佳工艺参数为:料液比1︰200(g/mL)、加酶量2%、酶解温度50℃、酶解时间5 h。在此条件下,螺旋藻细胞的破壁率为85.2%,与预测值84%接近。
Enzymatic hydrolysis was used to break Spriulina cell wall.The effects of solid-liquid radio,enzyme amount,temperature and hydrolysis time on the disruption were investigated by single factor tests and response surface methodology.The experiment results showed that the optimal condition for hydrolysis was:solid-to-liquid ratio of 1︰200(g/m L),enzyme amount of 2%,temperature of 50℃and process time of 5 h.The cell breaking rate was 85.2%under this condition,which perfectly matched the predicted value(84%).
