柱前衍生-高效液相色谱法检测测定沉香叶中γ-氨基丁酸的含量 被引量:2

Determination of the content of γ-aminobutyric acid in leaves of aquilaria sinensis(Lour.) spreng with pre-column derivatization-high performance liquid chromatography

摘要 目的建立测定沉香叶中γ-氨基丁酸的高效液相色谱方法。方法样品经过2,4-二硝基氟苯衍生化后,高效液相色谱法测定衍生物的含量。采用Kromasil C18色谱柱(150 mm×4.6 mm,5μm),流动相A为V(乙腈):V(超纯水)=1:1,流动相B为pH6.8含有3%四氢呋喃的磷酸盐缓冲液,A:B=35:65;检测波长为360 nm,柱温为27℃,流速为1.000 mL/min。结果γ-氨基丁酸衍生物在0.01~0.1 g/L范围内呈良好线性关系,r=0.9994;3批样品中γ-氨基丁酸平均含量为0.2983 mg/g。结论沉香叶中含有γ-氨基丁酸,所建立的含量测定方法可准确测定沉香叶中γ-氨基丁酸的含量。 Objective To establish an HPLC method for the determination ofγ-aminobutyric acid in leaves of aquilaria sinensis(Lour.)spreng.Methods Sample was prepared with 2,4-dinitrofluorobenzene derivatization and the content was determined with HPLC.In the HPLC analysis,the Kromasil C18 column(150 mm×4.6 mm,5μm)was used,the mobile phase A consisted of acetonitrile-water(1:1)while the mobile phase B was pH6.8 phosphate buffer with 3% tetrahydrofuran added(A:B=35:65),the UV detection wavelength was 360 nm and the column temperature was 27℃.Resultsγ-aminobutyric acid derivatives showed a good linear relationship in the range of 0.01-1.0 g/L,r=0.9994.The average content ofγ-aminobutyric acid in three batches of samples was 0.2983 mg/g.Conclusion The established method is simple,rapid and accurate.It can be used for the determination ofγ-aminobutyric acid in leaves of aquilaria sinensis(Lour.)spreng.
