小麦种子老化处理对发芽能力与蛋白组分的影响 被引量:4

摘要 以河南小麦百农4199为材料,高温、高湿老化处理后,对种子的发芽势、水溶蛋白、盐溶蛋白、醇溶蛋白和碱溶蛋白组分进行分析,研究小麦种子老化处理后发芽能力与种子贮藏蛋白组分变化之间的关系。结果表明:老化处理1 d后发芽势维持100%, 2~6 d缓慢下降, 7~8 d迅速降低,小麦种子失去发芽能力。老化处理1~4 d水溶蛋白20、23、35、60 ku组分,盐溶蛋白60 ku组分和醇溶蛋白80 ku组分均随着老化时间延长而增加;老化处理5~6 d水溶蛋白45、60 ku组分和醇溶蛋白100 ku组分消失,同期盐溶蛋白60 ku和醇溶蛋白80 ku出现新增组分;老化处理7~8 d各蛋白组分均表现条带模糊;碱溶蛋白在种子老化过程中没有明显条带增减变化。小麦种子老化过程中不同蛋白组分稳定性存在差异,老化处理5~6 d是种子发芽能力丧失和蛋白组分改变的转折期。 In order to elucidate the relationship between germination ability of wheat seeds and changes of seed storage protein components after aging treatment, the new wheat variety bainong 4199 in henan was studied by using artificial accelerated aging [temperature:(41±0.5) ℃;relative humidity 90%] with aging treatment 0, 1, 2, 3, 4, 5, 6, 7, 8 d. Germination potential and protein components in water, salt, alcohol and alkali solution of aged seeds were analyzed. The results showed that germination ability of wheat seeds was lost after 8 days of aging treatment, and germination maintained 100% at 1 day, decreased slowly from 2 days to 6 days, and decreased rapidly at 7 and 8 days after treatment. The changes of protein composition showed as follows: the contents of water-soluble protein 20, 23, 35 and 60 ku, salt-soluble protein 60 ku and alcohol-soluble protein 80 ku increased with the aging time during 1-4 days, and the components of 45, 60 and 100 ku of aqueous soluble protein disappeared after aging time during day 5-6, and new components of salt-soluble protein(60 ku) and alcohol-soluble protein(80 ku) appeared after aging time during 5-6 days, changes the protein components showed blurred bands after aging time during day 7-8. There were no obvious increases or decrease of alkali-soluble proteins during seed aging. The results showed that the stability of different protein components in wheat seed aging process was different. Aging treatment for 5-6 days is the transition period of seed germination loss and protein composition change. Our study provided a theoretical basis for identifying the characteristic markers of seed losing germination ability.
