摘要
目的采用超高效液相色谱法(UPLC)测定妇科再造丸中多指标成分及建立特征图谱的方法,为妇科再造丸的质量控制提供可靠依据。方法UPLC法,采用ACQUITY UPLC Hss T3(2.1×100mm,1.8μm)色谱柱,以0.1%甲酸水溶液(A)-乙腈(B)为流动相进行梯度洗脱;柱温:45℃;流速:0.3mL·min^-1;检测波长:280nm;进样量2μL。结果橙皮苷、黄芩苷分别在0.017~0.045,0.030~0.085μg内线性关系良好(r≥0.9995),平均回收率分别为99.71%(RSD 1.98%),102.27%(RSD 2.04%)。利用《中药色谱指纹图谱相似度评价系统2004A版》进行分析,共确定妇科再造丸UPLC特征图谱15个共有峰,通过混合对照比较确认其中2个指标性成分分别是橙皮苷(9号峰),黄芩苷(10号峰);对12批样品的特征图谱进行相似度分析,各批样品的相似度均在0.9以上。结论建立了妇科再造丸中指标成分测定及UPLC特征图谱,为妇科再造丸的质量评价提供科学依据。
OBJECTIVE To establish multiple index components determination method and characteristic spectrum method of FuKe ZaiZao Pills by UPLC,so as to provide reliable basis for the quality control.METHODS UPLC method was adopted.ACQUITY UPLC Hss T3(2.1×100mm,1.8μm)column was used,the mobile phase was 0.1%formic acid(A)-acetonitrile(B)with gradient elution.The flow rate was 0.3mL·min^-1,the column temperature was 45 degrers,the detection wavelengths was 280nm,and the injection volume was 2μL.RESULTS Hesperidin and baicalin had good linear relationship in the ranges of 0.017-0.045,0.030-0.085μg,respectively.The correlation coefficients were all more than 0.9995.The average recoveries were 99.71%(RSD 1.98%),102.27%(RSD 2.04%),respectively.Chinese Medicine Fingerprint Similarity Evaluation System 2004A edition was used for the analysis and totally 15 common peaks were identified.Compared with the mixed reference materials,two indicators were identified:hesperidin(peak 9)and baicalin(peak 10).The similarity of characteristic spectrums of 12 batches of samples was analyzed,and that of each batch was above 0.9.CONCLUSION The established multiple index components determination method and characteristic spectrum method by UPLC can be used for the quality control of FuKe ZaiZao Pills.
