宫颈癌外周血中的长链非编码RNA表达及其临床意义

摘要 目的探讨宫颈癌外周血中的长链非编码RNA(lncRNA)表达及其临床意义。方法选择病理学确诊的宫颈癌患者71例为病例组,选择同期在门诊进行常规体检的健康妇女71例为对照组,抽取2组入选者的外周血,采用基因芯片技术检测lncRNA表达情况,并采用实时荧光定量RCR进行验证分析。结果在芯片体系中,寡核苷酸芯片的外标、内标等阳性对照信号正常,阴性对照检测为阴性;漏点率不超过0.3%,平均背景值与噪音值较低;与对照组相比,病例组患者外周血中有差异表达基因12个,其中表达下调的基因有9个,下调倍数为(11.43±1.48);表达上调的基因有3个,上调倍数为(7.43±1.22)。随机挑选4个基因(KLF2、KLF6、KLF8、KLF3)进行实时荧光定量PCR验证,其检测结果与芯片检测的结果是一致的。结论lncRNAs基因芯片是检测宫颈癌外周血差异表达基因的一种高通量、高效、快速的检测手段,KLF2、KLF6、KLF8、KLF3可能是潜在的基因治疗靶点。 Objective To investigate the expression and clinical significance of lncRNA in peripheral blood of cervical cancer.Methods A total of 71 patients with cervical cancer diagnosed by pathology in a certain were selected as the case group,and the other 71 healthy women who were routinely examined in the outpatient clinic were selected as the control group.All the cases were extracted the peripheral blood and the expression of lncRNA were detected by gene chip technology,and the real-time fluorescence quantitative RCR were used for verification analysis.Results In the chip system,the positive control signals such as external standard and internal standard of the oligonucleotide chip were normal,and the negative control were negative;the leak rate were less than 0.3%,and the average background value and noise value were lower;there were 12 genes were differentially expressed in the peripheral blood of which 9 genes were down-regulated,the down-regulation multiple were 11.43±1.48,and there were 3 genes were up-regulated,and the up-regulation multiple were 7.43±1.22.Four genes(KLF2,KLF6,KLF8,KLF3)were randomly selected for real-time quantitative PCR,and the results were consistent with the results of the chip detection.Conclusion The lncRNAs gene chip is a high-throughput,high-efficiency and rapid detection method for detecting differentially expressed genes in peripheral blood of cervical cancer.KLF2,KLF6,KLF8 and KLF3 may be potential gene therapy targets.
